柞蚕蛹蛋白的提取及分子量测定的实验研究

摘  要:柞蚕蛹历史悠久,年产量丰富,富含蛋白优质,虽然已经应用于食品、医药、纺织等领域,但其工艺尚不成熟,工厂大批量生产难以保障其蛋白品质,无意间造成资源上的浪费。利用丙酮对柞蚕蛹进行脱脂,选用超声辅助碱水解法来提取脱脂蛹粉的蛋白,然后采用SDS-聚丙烯酰胺凝胶电泳法对蛹蛋白质提取液进行电泳实验,测定蛹蛋白的相对分子质量,测得其相对分子质量范围大部分为14.4~31.0 kDa,相对分子质量较小,可达到分子肽级别,有益于人体吸收,该方法可为柞蚕蛹蛋白在食品添加剂、医药、护肤品等领域的综合开发利用提供理论依据。The history of tussah pupae is long,with abundant annual production and high-quality protein.Although it has been applied in food,medicine,textile and other fields,its technology is not yet mature,and large-scale production in factories is difficult to guarantee its protein quality,inadvertently causing waste of resources.In this paper,acetone is used to degrease the tussah pupa,and ultrasonic assisted alkaline hydrolysis is used to extract the protein from the degreased pupa powder.Then SDS polyacrylamide gel electrophoresis is used to carry out electrophoresis experiments on the pupa protein extract,and the molecular weight of the pupa protein is measured.The molecular weight range is mostly 14.4~31.0 kDa,which is small and can reach the level of molecular peptide,which is beneficial to human absorption.This method can provide a theoretical basis for the comprehensive development and utilization of tussah pupa protein in food additives,medicine,skin care products and other fields.
