摘 要:目的探讨大蒜素对盐敏感性高血压(SSH)模型大鼠肠黏膜屏障及短链脂肪酸(SCFAs)的影响。方法本实验时间为2022年2—8月。将24只Dahl盐敏感大鼠随机分为正常盐组、高盐组和高盐+大蒜素组,每组8只。正常盐组大鼠给予0.3%氯化钠饲料喂养。高盐组和高盐+大蒜素组大鼠制备SSH模型,在此基础上,高盐+大蒜素组大鼠给予大蒜素16 mg/kg,口服。三组大鼠均干预4周。干预4周后,采用HE染色观察各组大鼠结肠组织病理学改变,采用Western blot法检测各组大鼠结肠组织Occludin、闭锁小带蛋白1(ZO-1)相对表达量,采用ELISA法检测各组大鼠血清炎症因子〔肿瘤坏死因子α(TNF-α)、白介素1β(IL-1β)、白介素6(IL-6)〕水平,采用液相色谱-串联质谱(LC-MS/MS)法检测各组大鼠血浆SCFAs(乙酸盐、丙酸盐、丁酸盐)水平,采用免疫组化染色法检测各组大鼠肾组织嗅觉受体(Olfr)78、G蛋白偶联受体(GPR)41表达水平,采用Western blot法检测各组大鼠肾组织Olfr78、GPR41蛋白相对表达量。结果HE染色结果显示:与正常盐组相比,高盐组结肠组织微绒毛大量减少、断裂,组织间排列疏松,间隙增大;与高盐组相比,高盐+大蒜素组结肠组织上皮结构相对完整,微绒毛排列紧密。干预4周后,高盐组大鼠结肠组织Occludin、ZO-1相对表达量低于正常盐组(P<0.05);干预4周后,高盐+大蒜素组大鼠结肠组织Occludin、ZO-1相对表达量高于高盐组(P<0.05)。干预4周后,高盐组大鼠血清TNF-α、IL-1β、IL-6水平高于正常盐组(P<0.05);干预4周后,高盐+大蒜素组大鼠血清TNF-α、IL-1β、IL-6水平低于高盐组(P<0.05)。干预4周后,高盐组大鼠血浆乙酸盐、丙酸盐、丁酸盐水平低于正常盐组(P<0.05);干预4周后,高盐+大蒜素组大鼠血浆乙酸盐、丙酸盐、丁酸盐水平高于高盐组(P<0.05)。干预4周后,高盐组大鼠肾组织Olfr78表达水平及其蛋白相对表达量高于正常盐组,GPR41表达水平Objective To investigate the effects of allicin on intestinal mucosal barrier and short-chain fatty acid(SCFAs)in salt-sensitive hypertension(SSH)model rats.Methods The experiment was conducted from February to August 2022.Twenty-four Dahl salt-sensitive rats were randomly divided into normal-salt group,high-salt group,and high-salt+allicin group,with 8 rats in each group.Rats in normal salt group were fed with 0.3%sodium chloride feed.The rats in the high-salt group and high-salt+allicin group were used to establish the SSH model.Additionally,the rats in the high-salt+allicin group received 16 mg/kg allicin orally.All groups were intervened for 4 weeks.After 4 weeks of intervention,HE staining was used to observe the histopathological changes of colonic tissue of rats in each group,the relative expression of Occludin,zonula occludens 1(ZO-1)in colon tissue of rats in each group was detected by Western blot method,the serum levels of inflammatory factors[tumor necrosis factorα(TNF-α),interleukin-1β(IL-1β)and interleukin-6(IL-6)]of rats in each group were detected by ELISA,the plasma levels of SCFAs(acetate,propionate,butyrate)of rats in each group were detected by liquid chromatography-tandem mass spectrometry(LC-MS/MS),the expression levels of olfactory receptor(Olfr)78 and G-protein coupled receptor(GPR)41 in renal tissue of rats in each group were detected by immunohistochemical staining,the relative expression levels of Olfr78 and GPR41 protein in renal tissues of rats in each group were detected by Western blot.Results HE staining results showed that compared with normal-salt group,microvilli of colon tissue in high-salt group were greatly reduced and broken,intertissue arrangement was loose,and space was enlarged;compared with the high-salt group,the epithelial structure of colon tissue in the high-salt+allicin group was relatively complete,and the microvilli were closely arranged.After 4 weeks of intervention,the relative expression of Occludin and ZO-1 in colon tissue of rats in high-salt group was lo
