摘  要:蛋白质组学前处理是蛋白质组学研究的关键,直接影响到后续分析的准确性和可靠性.文章综述了蛋白质组学前处理各环节的研究进展,包括样品制备、蛋白质提取、蛋白质消化、肽分级和肽纯化等.详细介绍了各种技术方法的原理、优缺点以及应用范围,阐述了采用新型蛋白酶组合的酶解方法及样本特异数据库的数据解析方法,并对未来的发展趋势进行了展望.The pretreatment of proteomics is the key to proteomics research and directly affects the accuracy and reliability of subsequent analyses.This article reviews the progress in all aspects of proteomics pretreatment,including the progress in sample preparation,protein extraction,protein digestion,peptide fractionation and peptide purification.It elaborates on the principles,advantages and disadvantages as well as the application ranges of various technical methods,expounds the enzymatic hydrolysis methods using new protease combinations and the data analysis methods for sample-specific databases,and also looks forward to the future development trends.
