摘 要:目的探究USP8调控TWIST1对卵巢癌细胞上皮间质转化的影响。方法将卵巢癌OVCAR3细胞和SKOV3细胞均随机分为si NC实验组、si USP8实验组和si TWIST1实验组。采用细胞集落实验分析OVCAR3和SKOV3细胞的增殖能力;采用Transwell实验方法分析OVCAR3和SKOV3细胞的侵袭能力;采用细胞划痕实验分析OVCAR3和SKOV3细胞的迁移能力;采用蛋白免疫印迹方法分析N-cadherin、Vimentin、USP8与TWIST1蛋白的表达。结果抑制USP8表达后,细胞集落实验显示,si USP8以及si TWIST1实验组的OVCAR3和SKOV3细胞增殖能力下降,差异均有统计学意义(P<0.05);Transwell实验显示,si USP8以及si TWIST1实验组的OVCAR3和SKOV3细胞侵袭数量下降,差异均有统计学意义(P<0.05);细胞划痕实验显示,si USP8以及si TWIST1实验组的OVCAR3和SKOV3细胞迁移率下降,差异均有统计学意义(P<0.05);蛋白免疫印迹实验结果显示,si USP8以及si TWIST1实验组的N-cadherin、Vimentin蛋白表达水平下降,si USP8实验组的USP8、TWIST1蛋白表达水平下降,差异均有统计学意义(P<0.05)。结论抑制USP8表达后,OVCAR3和SKOV3细胞的上皮间质转化过程受到抑制,该机制与USP8调节TWIST1密切相关。Objective To investigate the effect of USP8 regulating TWIST1 on epithelial mesenchymal transition(EMT)of ovarian cancer cells.Methods OVCAR3 and SKOV3 cells were randomly divided into si NC group,si USP8 group,and si TWIST1 group.Colony formation assay was used to analyze the proliferation of OVCAR3 and SKOV3 cells.Transwell assay was used to analyze the invasion ability of OVCAR3 and SKOV3 cells.The migration ability of OVCAR3 and SKOV3 cells was analyzed by cell scratch test.The protein expressions of N-cadherin,Vimentin,USP8 and TWIST1 were analyzed by Western blot.Results After inhibition of USP8 expression,colony formation assay showed that the proliferation ability of OVCAR3 and SKOV3 cells in the si USP8 and si TWIST1 groups was decreased significantly(P<0.05).Transwell assay showed that the invasion numbers of OVCAR3 and SKOV3 cells in the si USP8 and si TWIST1 groups were decreased significantly(P<0.05).Cell scratch test showed that the migration rates of OVCAR3 and SKOV3 cells in the si USP8 and si TWIST1 groups were decreased significantly(P<0.05).Western blot results showed that the expression levels of N-cadherin and Vimentin proteins in the si USP8 and si TWIST1 groups were decreased,and the expression levels of USP8 and TWIST1 proteins in the si USP8 group were decreased significantly(P<0.05).Conclusion Inhibition of USP8 expression inhibited the epithelial-mesenchymal transition in OVCAR3 cells and SKOV3 cells,and the mechanism is related to USP8 regulating TWIST1.
