香加皮三萜类化合物对食管癌大鼠模型免疫功能的影响及作用机制

摘  要:目的分析香加皮三萜类化合物对食管癌大鼠模型免疫功能的影响及作用机制。方法50只大鼠选取其中45只进行实验,10只作为对照组,其余35只建立食管癌大鼠模型,成功建模30只,随机分为药物对照组、模型组、研究组各10只。研究组给予香加皮三萜类化合物药液10 ml/kg;药物对照组给予大豆油药液10 ml/kg;对照组、模型组给予生理盐水10 ml/kg。检测各组免疫功能指标、炎性因子、滑膜细胞凋亡率、胸腺指数、热痛阈和脾脏指数水平、P53、IKb激酶抑制剂(IKK)-β、肿瘤坏死因子(TNF)-α、B细胞淋巴瘤(Bcl)-2相关X蛋白(Bax)、Bcl-2、核因子(NF)-κB蛋白相对表达量。结果与对照组相比,模型组、药物对照组CD4^(+)、热刺激缩足反应潜伏期(PWTL)、凋亡率(24、48、72 h)、Bax表达明显降低,CD4^(+)/CD25^(+)、胸腺指数、脾脏指数、P53、IKK-β、TNF-α、Bcl-2、NF-κB表达明显上升(P<0.05)。与模型组相比,研究组CD4^(+)/CD25^(+)、IL-10(15 w)、胸腺指数、脾脏指数、P53、IKK-β、TNF-α、Bcl-2、NF-κB表达明显降低,CD4^(+)、IL-2、IL-10(9 w)、凋亡率(24、48、72 h)、PWTL、Bax表达明显升高(P<0.05)。与药物对照组相比,研究组CD4^(+)、CD4^(+)/CD25^(+)(9 w)、IL-2、P53、IKK-β、TNF-α、Bcl-2、胸腺指数、脾脏指数明显降低,CD4^(+)/CD25^(+)(15 w)、IL-10、凋亡率(24、48、72 h)、PWTL、Bax表达明显升高(P<0.05)。结论香加皮三萜类化合物可改善食管癌大鼠免疫功能,降低炎性因子,促进滑膜细胞凋亡,其作用机制可能与P53、IKK-β、TNF-α、Bcl-2、Bax、NF-κB表达受到调节有关。Objective To analyze the influence and action mechanism of triterpenoids compound from Xianggapi on immune function in a rat model of esophageal cancer.Methods 45 out of 50 rats were selected for the experiment,10 rats were used as the control group,and the remaining 35 were used to establish the rat model of esophageal cancer.30 rats were successfully modeled,rats were randomly divided into drug control group,the model group,and the research group,with 10 rats in each group.The research group was given 10 ml/kg of Xianggapi triterpenoids compound solution;the drug control group was given 10 ml/kg of soybean oil solution;and the control and model groups were given 10 ml/kg of saline.The immune function indexes,inflammatory factors,synovial cell apoptosis rate,thymus index,heat pain threshold and splenic index levels,P53,IKb kinase inhibitor(IKK)-β,tumor necrosis factor(TNF)-α,B-cell lymphoma(Bcl)-2-associated X protein(Bax),Bcl-2,and relative expression of nuclear factor(NF)-κB protein in each group were detected.Results Compared with the control group,CD4^(+),latency of response to thermal stimulation of foot shrinkage(PWTL),cell apoptosis rate(24,48,72 h)and the expression of Bax were significantly decreased,CD4^(+)/CD25^(+),thymus index,splenic index,the expressions of P53,IKK-β,TNF-α,Bcl-2 and NF-κB were significantly increased in the model group and the drug control group(P<0.05).Compared with the model group,CD4^(+)/CD25^(+),IL-10(15 w),thymus index,splenic index,the expressions of P53,IKK-β,TNF-α,Bcl-2 and NF-κB were significantly decreased,CD4^(+),IL-2,IL-10(9 w),cell apoptosis rate(24,48,72 h),PWTL and the expression of Bax were significantly increased in the research group(P<0.05).Compared with the drug control group,CD4^(+),CD4^(+)/CD25^(+)(9 w),IL-2,P53,IKK-β,TNF-α,Bcl-2,thymus index,splenic index were significantly decreased,CD4^(+)/CD25^(+)(15 w),IL-10,cell apoptosis rate(24,48,72 h),PWTL and the expression of Bax were significantly increased in the research group(P<0.05).Conclusions The
