摘 要:随着人口老龄化加剧,心脑血管疾病已成为危害人类健康的常见病和主要死因之一。开发天然抗凝活性物质对防治心脑血管疾病有重大意义。方格星虫(Sipunculus nudus L.)是一种主要栖息于中国南方近海的底栖动物,其体壁富含多糖等生物活性物质,但目前还没有关于其抗凝血活性的研究报道。本文从方格星虫体壁中分离纯化了一种非中性多糖,结果表明,方格星虫多糖(SLP)的产率为5.76%,主要由Man、Rib、Gal和Glc组成,物质的量比为1∶2.83∶5.34∶6.95。SLP的数均分子量为9201 g/mol,聚合物分散性指数(PDI)为1.05,表明其为均一多糖。SEM和FT-IR分析显示SLP呈不规则片状结构,主要以α-吡喃糖构象存在。SLP表现出对APTT的延长作用,在200~400µg/mL时呈现量效关系,但800µg/mL时活性趋于饱和。目前,SLP的抗凝活性尚不及海参和硫酸软骨素等海洋硫酸化多糖,这与其硫酸基浓度低有关,也暗示了SLP可能具有不同于其他海洋多糖的抗凝血机制,值得进一步研究。综上,本研究为开发海洋动物来源的新型抗凝候选药物提供了新的候选物质及研究思路。With an aging population,cardiovascular and cerebrovascular diseases have become common illnesses and one of the main causes of death in humans.The development of natural anticoagulant bioactive compounds is of great significance for the prevention and treatment of these diseases.Sipunculus nudus L.is a marine benthic animal mainly inhabiting the offshore areas of southern China.Its body wall is rich in bioactive substances such as polysaccharides.However,no reports are available on its anticoagulant activity yet.Herein,a nonneutral polysaccharide was isolated and purified from the body wall of S.nudus L.The yield of S.nudus polysaccharide(SLP)was 5.76%.It mainly comprised Man,Rib,Gal,and Glc,with a molar ratio of 1∶2.83∶5.34∶6.95.The Mn of SLP was 9201 g/mol,and the polymer dispersity index(PDI)was 1.05,indicating its high uniformity.SEM and FT-IR analysis showed that SLP exhibited an irregular lamellar structure and mainly existed in the α-pyranose configuration.SLP exhibited a prolonging effect on APTT,showing a dose-effect relationship at 200-400μg/mL;however,the activity saturated at 800μg/mL.The anticoagulant activity of SLP is still inferior to that of chondroitin sulfate and marine sulfated polysaccharides such as those from sea cucumbers,which may be related to its low sulfate content.This finding also suggests that SLP may have a different anticoagulant mechanism from other marine polysaccharides,worthy of further research.In summary,this study provides new candidate substances and research avenues for developing novel anticoagulant drugs derived from marine animals.
