摘 要:目的基于生物信息学探讨核糖体蛋白8(RPS8)在骨软组织肉瘤(SARC)中的表达,并评估SARC患者预后的价值及其与免疫浸润的相关性。方法通过GEPIA数据库分析RPS8在SARC中的表达,通过GEPIA、Sangerbox数据库分析RPS8与肿瘤患者预后的相关性,通过Timer数据库分析RPS8与免疫细胞浸润的相关性,通过STRING数据库分析RPS8在SARC患者中与各种蛋白表达的相关性。结果肿瘤组织中RPS8表达水平高于正常组织,但差异无统计学意义(P>0.05);RPS8低表达组患者的生存期较高表达组显著延长,且PRS8高表达的男性患者预后最差(P<0.0001);RPS8与B细胞、CD8+T细胞、树突状细胞、中性粒细胞、巨噬细胞的浸润呈负相关,但与肿瘤纯度、CD4+T细胞无相关性;RPS8在SARC中的表达与TRMT基因家族、DNMT38、NOP2等RNA修饰基因密切相关,与VEGFB、TGFB1等免疫检查基因具有高度的相关性;TP53、ATRX、TTN是SARC中与RPS8表达相关度最高的前3项突变过程;且RPS8与RPS、RPL家族蛋白有显著的联系。结论在SARC患者中,RPS8的高表达与预后、免疫浸润密切相关,是SARC患者预后的危险因素,有望作为新的生物学标志物评估SARC患者的预后。Objective To explore the expression of ribosomal protein S8(RPS8)in bone and soft-tissue sarcoma(SARC)based on bioinformatics,and to evaluate the prognostic value of SARC patients and its correlation with immune infiltration.Methods The expression of RPS8 in SARC was analyzed using the GEPIA database.The correlation between RPS8 and the prognosis of tumor patients was analyzed using the GEPIA and Sangerbox databases.The correlation between RPS8 and immune cell infiltration was analyzed using the Timer database.The correlation between RPS8 and various protein expressions in SARC patients was analyzed using the STRING database.Results The expression level of RPS8 in tumor tissues was higher than that in normal tissues,but the difference was not statistically significant(P>0.05).The survival time of patients in the RPS8 low expression group was significantly longer than that in the high expression group,and the prognosis of male patients with high PRS8 expression was the worst(P<0.0001).RPS8 was negatively correlated with the infiltration of B cells,CD8+T cells,dendritic cells,neutrophils and macrophages,but not with tumor purity and CD4+T cells.The expression of RPS8 in SARC was closely related to RNA modification genes such as TRMT gene family,DNMT38 and NOP2,and was highly correlated with immune examination genes such as VEGFB and TGFB1.TP53,ATRX and TTN were the first three mutation processes with the highest correlation with RPS8 expression in SARC.RPS8 was significantly associated with RPS and RPL family proteins.Conclusion In SARC patients,the high expression of RPS8 is closely related to prognosis and immune infiltration,and is a risk factor for the prognosis of SARC patients.It is expected to be used as a new biomarker to evaluate the prognosis of SARC patients.
