摘 要:为探究汉氏葡糖醋杆菌(Komagataeibacter hansenii,K.hansenii)HDM1-3所包含的GGDEF结构域对合成环二鸟苷酸(cyclic diguanylate monophosphate,c-di-GMP)的作用,挖掘c-di-GMP对该菌种产细菌纤维素(Bacterial cellulose,BC)的作用,采用生物信息学方法系统分析了K.hansenii HDM1-3菌株的基因组,发现K.hansenii HDM1-3菌株中共有8个基因含有GGDEF-EAL结构域。利用Hmmer软件、Pfam数据库、SMART(Simple Modular Architecture Research Tool)平台、MEME Suite平台、MEGA11(Molecular Evolutionary Genetics Analysis 11)软件、ClustalW平台、Swiss-model平台、NCBI(National Center for Biotechnology Information)平台、InterPro平台及BLAST(Basic Local Alignment Search Tool)平台对该菌株所含有GGDEF-EAL结构域的功能进行了分析,并深入探究了该结构域对细菌纤维素产量的影响。功能预测分析表明,K.hansenii HDM1-3中含有GGDEF-EAL结构域的8个基因均参与c-di-GMP的合成、降解及信号传导。In order to investigate the role of GGDEF domain in the synthesis of cycli diguanylate monophosphate(c-di-GMP)in Komagataeibacter hansenii(K.hansenii)HDM1-3,and explore the effects of c-di-GMP on bacterial cellulose(BC)production in this strain,a systematic analysis of the genome of K.hansenii HDM1-3 was conducted using bioinformatics methods.It has been found that there are a total of 8 genes containing the GGDEF-EAL domain in the K.hansenii HDM1-3.The functionality of these GGDEF-EAL domains was analyzed and their impact on bacterial cellulose production was explored using Hmmer software,Pfam database,SMART(Simple Modular Architecture Research Tool)platform,MEME Suite platform,MEGA11(Molecular Evolutionary Genetics Analysis 11)software,ClustalW platform,swiss-model platform,NCBI(National Center for Biotechnology Information)platform,InterPro platform,and BLAST(Basic Local Alignment Search Tool)platform.Functional prediction analysis indicates that the eight genes containing GGDEF-EAL domains in K.hansenii HDM1-3 are involved in the synthesis,degradation,and signaling of c-di-GMP.
