醒神涤肠汤对A型肝性脑病大鼠的防治作用

摘　　要：目的研究醒神涤肠汤对A型肝性脑病大鼠的防治作用。方法以雄性SPF级大鼠为实验对象,将其按随机数字表法分成正常组、模型组、对照组、药物组,各组8只。正常组、模型组10 ml/(kg•d)生理盐水灌胃,对照组予以乳果糖10 ml/(kg•d)灌胃,药物组为10.625 g/(kg•d)醒神涤肠汤灌胃,共7 d,每日2次。在第6天时开始进行造模,模型组、对照组、药物组大鼠均腹腔内注射硫代乙酰胺(TAA)300 mg/(kg•d),连续给予2 d,正常组用等量生理盐水经腹腔注射。观察大鼠并对其脑功能进行评分;测定ALT、AST、ALP、TBIL和肝脏病理切片反映组织损伤;检测血氨及结肠pH值。结果与模型组相比,药物组大鼠的脑功能评分显著增加,生化指标ALT、AST、ALP和TBIL显著降低(P<0.05);血氨和结肠pH值显著降低(P<0.05);大鼠肝组织病理学损伤明显改善。结论醒神涤肠汤可通过改善模型大鼠的脑功能,降低体内氨的含量,改善肝脏的损伤及肠pH值等多方面发挥治疗肝性脑病的作用。Objective In order to study the preventive and therapeutic effects of Xingshen Dichang Decoction on hepatic encephalopathy in rats.Methods Male SPF rats were divided into normal group,model group,control group and drug group according to random number table,with 8 rats in each group.Normal group and model group were daily gavage-fed with 10 ml/(kg•d)physiological saline,control group was gavage-fed with 10 ml/(kg•d)lactulose,and drug group was gavage-fed with 10.625 g/(kg•d)Xingshen Ditchang Decoction,for 7 consecutive days,twice a day.On day 6,the model was constructed,and Andrats in model group,control group and drug group were given intraperitoneally injection of thioacetamide(TAA)at 300 mg/(kg•d)for 2 consecutive days,normal group received an injection of the same quantity of normal saline.The rats were observed and their brain function was scored.ALT,AST,ALP,TBIL and pathological sections of liver were determined to reflect tissue injury.Blood ammonia and colon pH were detected.Results Compared with the model group,rats in the drug group showed a significant increase in brain function scores and a significant decrease in the biochemical indexes ALT,AST,ALP,and TBIL(P<0.05),blood ammonia and colon pH were significantly reduced(P<0.05),and hepatic histopathological damage in the rats was significantly ameliorated.Conclusions Xingshen Ditchang Decoction can play a role in treating hepatic encephalopathy by improving the brain function of the model rats,reducing the content of ammonia in the body,and improving liverand intestinal pH damage in multiple aspects.
