摘 要:目的:筛选胃癌铁死亡发生发展中的关键基因,并分析其功能和肿瘤免疫浸润的关系。方法:从TCGA数据库下载胃癌基因表达谱,用R软件筛选胃癌组织与正常组织样本间的差异表达基因(DEGs)并与铁死亡相关基因取交集。构建蛋白相互作用网络(PPI)筛选胃癌铁死亡进展的关键基因,通过R软件对铁死亡关键基因进行基因集富集分析(GSEA)。利用TIMER数据库分析关键基因与免疫浸润细胞的相关性。结果:共筛选出202个铁死亡相关的DEGs。通过PPI分析得到3个铁死亡关键基因,分别为周期蛋白依赖性激酶1(CDK1)、细胞周期蛋白依赖性激酶抑制剂2A(CDKN2A)和Zeste增强子同源物2(EZH2)。关键基因富集到二价无机阳离子稳态及脂质代谢等通路。免疫浸润分析得出,关键基因的表达与多种免疫细胞的浸润相关。结论:CDK1、CDKN2A和EZH2为胃癌铁死亡发生发展中的关键基因,并与胃癌的免疫浸润有关。Objective:To screen for key genes involved in the occurrence and development of ferroptosis in gastric cancer and to analyze their functions and relationship with tumor immune infiltration.Methods:Gene expression profiles of gastric cancer are downloaded from the TCGA database.Differentially expressed genes(DEGs)between gastric cancer tissues and normal tissues are screened using R software,and the intersection with ferroptosis-related genes is taken.A protein-protein interaction(PPI)network is constructed to identify key genes in the progression of ferroptosis in gastric cancer.Gene set enrichment analysis(GSEA)for key ferroptosis genes is conducted using R software.The TIMER database is utilized to analyze the correlation between key genes and immune infiltrating cells.Results:A total of 202 ferroptosis-related DEGs are identified.Three key ferroptosis genes were obtained through PPI analysis,namely cyclindependent kinase 1(CDK1),cyclin-dependent kinase inhibitor 2A(CDKN2A)and Zeste enhancer homolog 2(EZH2).GSEA for these key genes reveal pathways related to bivalent inorganic cation homeostasis,and lipid metabolism.Immune infiltration analysis indicate that the expression of key genes is associated with the infiltration of various immune cells.Conclusion:CDK1,CDKN2A,and EZH2 are key genes involved in the occurrence and development of ferroptosis in gastric cancer,and they are associated with immune infiltration in gastric cancer.
