摘 要:为优化苦苣多糖的提取工艺,分析苦苣多糖的理化特性与抗氧化活性,本文以苦苣为原料,以多糖得率为评价指标,在单因素实验的基础上采用响应面实验优化苦苣多糖的提取工艺,同时对优化后的多糖进行羟自由基清除实验、DPPH自由基清除实验和超氧阴离子自由基清除实验评价苦苣多糖的抗氧化活性。结果表明,苦苣多糖的最佳提取工艺为提取时间30 min、提取温度61℃、料液比1∶31(g∶mL),在此条件下,苦苣多糖的得率为7.16%,总糖含量为68.94%、糖醛酸含量为13.01%、总蛋白含量为2.11%。当苦苣多糖浓度为10 mg/mL时,羟自由基清除率达80.8%,DPPH自由基清除率为76.5%,超氧阴离子自由基清除率为78.0%。In order to optimize the extraction process of polysaccharides from Sonchus oleraceus L.and analyze the physicochemical properties and antioxidant activities of polysaccharides from Sonchus oleraceus L.,response surface experiment is used to optimize the extraction process of polysaccharides from Sonchus oleraceus L.on the basis of single factor experiment,with Sonchus oleraceus L.as raw material and polysaccharide yield as evaluation index.At the same time,the optimized polysaccharides are subjected to hydroxyl radical scavenging experiment,DPPH radical scavenging experiment and superoxide anion radical scavenging experiment.The results show that the optimal extraction process are as follows:extraction time 30 min,extraction temperature 61℃,solid-liquid ratio 1∶31(g∶mL).Under these conditions,the yield of polysaccharides was 7.16%,the content of total sugar was 68.94%,the content of glucuronic acid is 13.01%and the content of total protein is 2.11%.When the concentration of polysaccharide is 10 mg/mL,the scavenging rates of hydroxyl radical,DPPH radical and superoxide anion radical are 80.8%,76.5%and 78.0%respectively.
