摘 要:以紫薇(Lagerstroemia indica)品种‘建民红’的半同胞家系后代中的红、粉、白3种不同花色子代作为试材,克隆获取LiUFGT1基因的全长,对其进行生物信息学和表达分析,探究UFGT基因在紫薇不同花色中的作用,以期为进一步研究紫薇花色调控机制及其分子育种提供参考依据。结果表明:紫薇LiUFGT1基因的CDS区为1203 bp,共编码400个氨基酸,具有UDPGT保守结构域;其蛋白分子质量约为44.05 kD,等电点(pI)为5.51,属于不稳定的亲水性蛋白。系统进化树分析显示LiUFGT1与石榴PgUFGT基因亲缘关系最近,氨基酸相似性达93%。花青苷含量检测表明红花子代(R)在盛花期和末花期的花青苷含量较高,粉花子代(P)在花苞期、盛花期、末花期等3个花发育过程中花青苷含量逐渐上升;而白花子代(W)的花青苷含量低,总体呈现下降趋势。qRT-PCR分析显示LiUFGT1在R、P、W的不同花发育时期的表达趋势与花青苷含量变化一致,表明LiUFGT1基因的表达与花青苷呈正相关。Taking red,pink,and white offspring from half sibling families of the Lagerstroemia indica variety‘Jianminhong’as test materials,the full-length LiUFGT1 gene was cloned and analyzed for bioinformatics and expression to explore the role of the UFGT gene in different flower colors of Lagerstroemia indica,and in order to provide reference for further research on the regulation mechanism of flower color and molecular breeding of L.indica.The results showed that the CDS region of the LiUFGT1 gene in Lagerstroemia indica was 1203 bp,encoding a total of 400 amino acids and possessing a UDPGT conserved domain.The protein molecular weight was about 44.05 kD,with an isoelectric point(pI)of 5.051,and it belonged to an unstable hydrophilic protein.The phylogenetic tree analysis showed the closest genetic relationship with the pomegranate PgUFGT gene,with an amino acid similarity of 93%.The detection of anthocyanin content showed that the red flower offspring(R)had higher anthocyanin content during the peak and final flowering stages,while the pink flower offspring(P)gradually increased in anthocyanin content during the three flower development stages of bud,peak,and final flowering;the content of anthocyanins in the offspring of white flowers(W)was low,showing an overall downward trend.qRT-PCR analysis showed that the expression trend of LiUFGT1 at different flower development stages of R,P,and W was consistent with the changes in anthocyanin content.The results showed that the expression of LiUFGT1 gene was positively correlated with anthocyanins.
