摘  要:目的 研究溃疡性结肠炎(UC)模型大鼠的结肠组织代谢物质差异,并探索UC疾病机制其相关代谢通路及代谢物质研究。方法 采用超高效液相色谱-四极杆-飞行时间串联质谱法(UPLC-Q-TOF-MS)技术进行代谢组学分析,根据Pathway公共数据库,对鉴定出的代谢物通过MetaboAnalyst 5.0进行KEGG代谢通路注释分析。结果 与UC疾病密切相关的8条代谢通路分别是Pentose phosphate pathway、Galactose metabolism、Alanine, aspartate and glutamate metabolism、Arginine and proline metabolism、beta-Alanine metabolism、Arachidonic acid metabolism、Fatty acid metabolism、Fatty acid biosynthesis;通路中相关联10种物质分别为丙二酸、2-脱氧核糖1-磷酸、(±)5,6-二羟基-8Z,11Z,14Z-二十碳三烯酸、L-天冬酰胺、4-胍基丁酸、5-氨基戊酸、胍基乙酸、N-乙酰基-D-半乳糖胺、delta-12-Prostaglandin J2。结论 溃疡性结肠炎发展过程中肠道长期处于长期的氧化应激状态致使肠道黏膜屏障受损,而损伤的肠道黏膜对氨基酸、糖类等能量物质代谢发生障碍。Objective To investigate the differences in colonic tissue metabolites in a rat model of ulcerative colitis(UC)and explore the associated metabolic pathways and metabolites underlying UC pathogenesis.Methods Metabolomic analysis was performed using ultra-performance liquid chromatography quadrupole time-of-flight mass spectrometry(UPLC-Q-TOF-MS).Identified metabolites were annotated and analyzed for KEGG metabolic pathways via MetaboAnalyst 5.0 based on the KEGG Pathway database.Results Eight metabolic pathways closely linked to UC were identified:the pentose phosphate pathway,galactose metabolism,alanine/aspartate/glutamate metabolism,arginine/proline metabolism,beta-alanine metabolism,arachidonic acid metabolism,fatty acid metabolism,and fatty acid biosynthesis.Ten associated metabolites were determined:malonic acid,2-deoxyribose 1-phosphate,(±)5,6-dihydroxy-8Z,11Z,14Z-eicosatrienoic acid,L-asparagine,4-guanidinobutyric acid,5-aminovaleric acid,guanidinoacetic acid,N-acetyl-D-galactosamine,and delta-12-prostaglandin J2.Conclusion In Ulcerative Colitis development,long-term intestinal oxidative stress impairs the mucosal barrier,disrupting the metabolism of energy substances like amino acids and carbohydrates.
