摘 要:目的:研究土牛膝提取物对α-葡萄糖苷酶和二肽基肽酶-4(DPP-4)的抑制作用,探究其降糖作用机制。方法:采用系统溶剂提取法对土牛膝的根、茎、叶进行提取,以α-葡萄糖苷酶和DPP-4为靶点,建立酶活抑制模型,检测土牛膝提取物对酶的抑制作用。结果:土牛膝不同提取物对α-葡萄糖苷酶均有不同程度的抑制作用,其中叶的乙酸乙酯提取物对α-葡萄糖苷酶显示出强抑制活性,IC50为0.18μg/mL。土牛膝不同提取物苷酶;DPP-4;降对DPP-4也有不同程度的抑制作用,其中叶的二氯甲烷提取物对DPP-4显示出强抑制活性,IC50为44.57μg/mL。结论:土牛膝提取物对这两种酶均有不同程度的抑制作用。Objective:To study the inhibitory effects of Achyranthes aspera L.(AL)onα-glucosidase and dipeptidyl peptidase-4(DPP-4),and explore its mechanism of hypoglycemic action.Methods:The root,stem and leaf of AL were extracted by systematic solvent extraction method.Takingα-glucosidase and DPP-4 as targets,the inhibitory activity of AL extracts onα-glucosidase was evaluated by establishing enzyme inhibition model.Results:The root,stem and leaf extracts of AL showed different degrees of inhibition onα-glucosidase.The IC50 of all the ethyl acetate extracts onα-glucosidase was about 15μg/mL,showing good inhibitory activity.The ethyl acetate extracts of AL showed strong inhibitory activity onα-glucosidase.IC50 was 0.18μg/mL.Different extracts of AL also had different inhibitory effects on DPP-4,and the dichloromethane extract of AL showed strong inhibitory activity on DPP-4 with IC50 of 44.57μg/mL.Conclusion:The extract of AL has different inhibitory effects onα-glucosidase and dipeptidyl peptidase-4.
