摘  要:为探讨瑶药“青九牛”内生真菌Alternaria burnsii KJT-2的化学成分及其抗菌作用,本文对内生真菌Alternaria burnsii KJT-2进行发酵培养并获得其次级代谢产物,通过测定其化学成分、抑菌圈直径、最低抑菌浓度(MCI)、菌悬液的电导率、细菌细胞内物质外漏情况及β-半乳糖苷酶活性进行抗菌作用评价。研究结果表明内生真菌代谢产物中含有有机酸、苯丙素、酯类、生物碱、萜类等多种成分,其对金黄色葡萄球菌及链球菌有抑制活性,抑菌圈直径分别为20.30±0.8 mm和21.30±0.90 mm,MIC值分别为7.81 mg·mL^(-1)和31.25 mg·mL^(-1)。机制研究表明内生菌代谢产物能够改变细菌的细胞膜的通透性,导致细胞内的物质外漏,电导率和β-半乳糖苷酶活力升高,最终抑制细菌生长。本研究表明瑶药“青九牛”内生真菌Alternaria burnsii KJT-2代谢产物具有抗菌活性,为瑶药“青九牛”内生真菌资源的开发利用及活性化合物的发现提供了一定的理论基础。To investigate the chemical composition and antibacterial effect of Alternaria burnsii KJT-2,an endophytic fungus isolated from Yao medicine“Qingjiuniu”,the fungi were cultivated through fermentation to obtain secondary metabolites.The antibacterial activity was evaluated by measuring the chemical composition,inhibition zone diameter,minimum inhibitory concentration(MIC),electrical conductivity of bacterial suspensions,leakage of intracellular substances,andβ-galactosidase activity.The research results indicate that the metabolites of the endophytic fungus contain various components such as organic acids,phenylpropanoids,esters,alkaloids,terpenoids,and others.These metabolites exhibit inhibitory activity against Staphylococcus aureus and Streptococcus,with inhibition zone diameters of 20.30±0.8 mm and 21.30±0.90 mm,respectively,and MIC values of 7.81 and 31.25 mg·mL^(-1),respectively.Mechanism studies reveal that the metabolites of the endophytic fungus can alter the permeability of bacterial cell membranes,leading to the leakage of intracellular substances,increased electrical conductivity,and elevatedβ-galactosidase activity,ultimately inhibiting bacterial growth.This study demonstrates that the metabolites of Alternaria burnsii KJT-2,an endophytic fungus from the Yao medicine“Qingjiuniu”,possess antibacterial activity,providing a theoretical basis for the exploitation and utilization of this endophytic fungal resource and the discovery of active compounds.
