摘  要:目的分析黄芪总皂苷对实验性自身免疫性脑脊髓炎(experimental autoimmune encephalomyelitis,EAE)小鼠细胞因子表达的影响。方法于2021年10月—2022年5月选取研究所动物实验中心24只C57BL/6雌性小鼠,根据随机数字表法将其平均分为三组,EAE组注射磷酸缓冲盐溶液(phosphate buffer saline,PBS),低剂量组和高剂量组分别注射黄芪总皂苷溶液40 mg/(kg·d)和80 mg/(kg·d)。记录对比各组体质量、临床症状评分以及炎症因子和神经营养因子水平。结果免疫28 d时,低剂量组和高剂量组体质量均高出EAE组,且高剂量组高出低剂量组(P=0.042);免疫14 d和28 d时,低剂量组和高剂量组症状评分低于EAE组,且高剂量组低于低剂量组(P=0.026);免疫28 d后,AST治疗组白细胞介素-10(Interleukin-10,IL-10)表达水平高出EAE组,白细胞介素1β(Interleukin-1 beta protein,IL-β)表达水平显著低于EAE组(P<0.001),AST治疗组的BDNF表达水平和GAP-43蛋白含量较EAE组显著上升(P<0.001)。结论黄芪总皂苷治疗能够减缓小鼠体质量下降情况和临床症状,且高剂量治疗效果更加显著,可能是由于其对细胞因子及神经组织生长情况起到调节作用。Objective To analyze the effects of total astragalus saponins on cytokine expression in experimental autoimmune encephalomyelitis(EAE)mice.Methods From October 2021 to May 2022,24 C57BL/6 female mice in the animal experimental Center of the Institute were selected and divided into three groups by random number table method.The EAE group was injected with phosphate buffer saline(PBS).Low dose group and high dose group were injected with Astragalus saponins solution 40 and 80mg/kg/d,respectively.Body mass,clinical scores and levels of inflammatory factors and neurotrophic factors were recorded and compared.Results After 28 days of immunization,the body weight of low dose group and high dose group was higher than that of EAE group,and high dose group was higher than low dose group(P=0.042).Symptom scores in the low-dose and high-dose groups were lower than those in the EAE group,and those in the high-dose group were lower than those in the low-dose group(P=0.026).After 28 days of immunization,the expression of Interleukin-10(IL-10)in the AST treatment group was higher than that in the EAE group,and the expression of interleukin-1 beta protein(IL-β)in the AST treatment group was significantly lower than that in the EAE group(P<0.001).BDNF and GAP-43 proteins in AST treatment group were significantly higher than those in EAE group(P<0.001).Conclusion Astragalus saponins treatment can reduce the decline of body mass and clinical symptoms in mice,and the effect of high dose treatment is more significant,which may be due to its regulation of cytokines and nerve tissu e growth.
