摘  要:目的 探究桑色素(Morin)对支气管哮喘(BA)小鼠炎症反应的影响及其机制。方法 通过卵清蛋白/氢氧化铝悬液诱导BALB/c小鼠建立BA模型,并随机分为模型组、Morin低剂量组(Morin-30,30 mg/kg Morin)、Morin中剂量组(Morin-90,90 mg/kg Morin)、Morin高剂量组(Morin-180,180 mg/kg Morin)和Morin-180+鸦胆子苦醇组[180 mg/kg Morin+4 mg/kg核红细胞相关因子2(Nrf2)抑制剂brusatol],每组15只;另选取15只健康小鼠作为对照组。Morin低、中、高剂量组和Morin-180+鸦胆子苦醇组给药14 d后收集小鼠血液、支气管肺泡灌洗液和肺组织,分别通过酶联免疫吸附测定、流式细胞仪、迪夫快速(Diff-Quik)染色、比色法、苏木素伊红(HE)染色、免疫组织化学法和蛋白质印迹法检测免疫细胞、炎症因子[肿瘤坏死因子(TNF)-α、干扰素(IFN)-γ、白介素(IL)-4和IL-13]水平、氧化应激因子、肺组织病理变化及Nrf2/血红素加氧酶-1(HO-1)通路蛋白表达水平。结果 与对照组相比,模型组小鼠出现气道阻塞,且肺组织出现病理性损伤,TNF-α、IFN-γ、IL-4和IL-13水平及炎症细胞数量升高(P<0.05),Nrf2、HO-1蛋白表达降低(P<0.05);与模型组相比,Morin治疗组小鼠TNF-α、IFN-γ、IL-4和IL-13水平及炎症细胞数量下降(P<0.05),Nrf2、HO-1蛋白表达上升(P<0.05),肺组织损伤和炎症反应减轻;与Morin-180组相比,Morin-180+brusatol组小鼠TNF-α、IFN-γ、IL-4和IL-13水平及炎症细胞数量升高(P<0.05),Nrf2、HO-1蛋白表达降低(P<0.05),肺组织损伤和炎症反应加剧。结论 Morin可抑制BA小鼠炎症反应,其作用机制与Nrf2/HO-1信号通路有关。Objective To explore the effect of Morin on the inflammatory response in mice with bronchial asthma(BA)and its mechanism.Methods Mouse models of BA were established in BALB/c mice induced by ovalbumin/aluminum hydroxide suspension.And they were assigned into a model group,a low-dose Morin group(Morin-30,30 mg/kg),a medium-dose Morin group(Morin-90,90 mg/kg),a high-dose Morin group(Morin-180,180 mg/kg),and a Morin-180+bruceopitol group[180 mg/kg Morin+4 mg/kg nuclear factor erythroid 2-related factor 2(Nrf2)inhibitor brusatol],with 15 mice in each group.And 15 healthy mice were included in the control group.Mice in each group received corresponding treatment for 14 d.Samples of blood,bronchoalveolar lavage fluid and lung tissue were collected.The immune cells,inflammatory factors[tumor necrosis factor(TNF)-α,interferon(IFN)-γ,interleukin(IL)-4 and IL-13]levels,oxidative stress factors,lung tissue pathological changes,and Nrf2/heme oxygenase-1(HO-1)pathway protein expression levels were detected by enzyme-linked immunosorbent assay,flow cytometry,Diff-Quik staining,colorimetry,hematoxylin and eosin(HE)staining,immunohistochemistry staining,and Western blot,respectively.Results Compared with the control group,the model group exhibited airway obstruction,pathological damage to lung tissue,increased TNF-α,IFN-γ,IL-4 and IL-13 levels,increased number of inflammatory cells,but decreased expression of Nrf2 and HO-1 proteins(P<0.05).Compared with the model group,the Morin treatment groups showed decreased TNF-α,IFN-γ,IL-4 and IL-13 levels,decreased number of inflammatory cells,but increased expression of Nrf2 and HO-1 proteins(P<0.05),as well as improved lung tissue damage and reduced inflammation.Compared with the Morin-180 group,the Morin-180+brusatol group exhibited increased TNF-α,IFN-γ,IL-4 and IL-13 levels,increased number of inflammatory cells,but decreased expression of Nrf2 and HO-1 proteins(P<0.05),as well as worse lung tissue damage and aggravated inflammation.Conclusion Morin inhibits the inflammatory
