摘 要:【目的】为了帮助养殖场确立一种快速筛选消毒药的检测方法。【方法】选择过硫酸氢钾、医用碘伏、84消毒液、二溴海因四种常见的养殖场消毒药,以大肠杆菌为指示菌,采用点种法对四种消毒药在不同浓度下对大肠杆菌的抑制作用进行了研究,并使用试纸片法以及牛津杯法对点种法试验结果的准确性进行了验证。【结果】点种法试验结果表明,在10^(-1)浓度时,四种消毒药对大肠杆菌均有抑制作用;在10^(-3)浓度时,四种消毒药对大肠杆菌均没有抑制作用;延伸试验结果表明,在10^(-1)浓度时,四种消毒药均完全杀灭了大肠杆菌,而在10^(-3)时,四种消毒药对大肠杆菌则完全没有作用;牛津杯法抑菌试验以及试纸片法抑菌试验的结果在10^(-1)浓度时以及10^(-3)浓度时表现出来对大肠杆菌的杀灭作用与点种法试验的结果高度相似。【结论】说明了点种法相比较于传统的牛津杯法以及试纸片法更加高效;同时,通过常规的试纸片法以及牛津杯法对点种法试验也验证了点种法的准确性。[Objective]The study aimed to help farms to establish a rapid screening of disinfectants detection method.[Method]Four common farm disinfectants,potassium hydrogen persulfate,medical iodophor,84 disinfectant and dibromohydantoin,were selected,and Escherichia coli was used as indicator bacteria.The inhibitory effects of four disinfectants on Escherichia coli at different concentrations were studied by dot seed method.The accuracy of the experimental results of the dot method was verified by test strip method and Oxford cup method.[Result]The results of spot method showed that all the four disinfectants had inhibitory effects on Escherichia coli at 10^(-1) concentration,and none of the four disinfectants had inhibitory effects on Escherichia coli at 10^(-3) concentration;the results of extension experiment showed that all four disinfectants killed Escherichia coli completely at the concentration of 10^(-1),while at 10^(-3),the four disinfectants had no effect on Escherichia coli;the results of Oxford cup method and test strip method showed that the bactericidal effects on Escherichia coli at 10^(-1) concentration and 10^(-3) concentration were highly similar to that of spot method.[Conclusion]This consistency highlights the enhanced efficiency of the spotting method when compared to that of the traditional methods like the Oxford cup and test strip methods.Moreover,the accuracy of the spotting method was further validated by the conventional test strip and Oxford cup methods used in conjunction with the spotting method experiments,thereby affirming its reliability and suitability for disinfectant detection.
