摘 要:目的:探讨儿茶素的抗变应性鼻炎作用机制。方法:采用卵白蛋白制备AR大鼠模型;造模成功后,实验分为模型组,低、中、高剂量儿茶素[100、200、400 mg/(kg·d)]组、西替利嗪组[20 mg/(kg·d)],每组灌胃给药,连续给药14 d。采用叠加量化计分法评价变应性鼻炎大鼠鼻部行为学症状改变;运用伊文思蓝法、变色素2R染色法和甲苯胺蓝染色法分别检测鼻黏膜血管通透性、嗜酸性粒细胞和肥大细胞的数量;采用免疫组织化学法检测鼻黏膜组胺受体(Histamine H1 receptor, H1R)表达,ELISA法检测血清IL-4、IL-33、组胺水平。结果:与模型组比较,儿茶素和西替利嗪组大鼠抓绕鼻、打喷嚏和流清涕等鼻部行为学症状显著缓解(P<0.05或P<0.01);鼻黏膜血管通透性、嗜酸性粒细胞、肥大细胞数量、H1R表达水平和血清IL-4、IL-33、组胺水平显著下降(P<0.05或P<0.01),且儿茶素的作用效果呈一定的剂量依赖性。高剂量儿茶素对AR大鼠的疗效优于西替利嗪。结论:儿茶素可能通过下调组胺发挥抗变应性鼻炎药理效应,并可通过降低血清IL-33、IL-4水平减轻组胺引起的炎症反应。Objective:To investigation the mechanism of Catechins in anti allergic rhinitis.Methods:Allergic rhinitis(AR)rats models were prepared using ovalbumin.After successful modeling,the experiment was divided into a model group,low,medium,and high-dose Catechins[100,200,400 mg/(kg·d)]group,and Cetirizine group[20 mg/(kg·d)],each group was orally administered for 14 consecutive days.Using the stacked quantitative scoring method to evaluate the changes in nasal behavioral symptoms in rats with allergic rhinitis.The vascular permeability,eosinophil count,and mast cell count of nasal mucosa,respectively were detected by the evans blue staining,chromotrope 2R staining,and toluidine blue staining.The expression of histamine H1 receptor(H1R)in nasal mucosa was detected by immunohistochemical method,and the levels of IL-4,IL-33,Histamine in serum was detected by ELISA method.Results:Compared to the model group,rats in the Catechin and Cetirizine groups exhibited significant relief in nasal behavioral symptoms,including nose scratching,sneezing,and rhinorrhea(P<0.05 or P<0.01);Nasal mucosal vascular permeability,eosinophil count,mast cell count,H1R expression level,as well as serum levels of IL-4,IL-33,and histamine,all significantly decreased(P<0.05 or P<0.01),and the effect of Catechins showed a certain dose-dependent.The therapeutic effect of high-dose Catechins on AR rats was superior to that of Cetirizine.Conclusion:Catechins may exert pharmacological effects against allergic rhinitis by down-regulating histamine,and alleviate the inflammatory responses induced by histamine by reducing serum levels of IL-33 and IL-4.
