摘 要:试验旨在研究丁酸梭菌对犊牛肠道致病菌(金黄色葡萄球菌、大肠杆菌、沙门氏菌)的抑制作用和抗氧化性能。采用牛津杯法进行抑菌试验,以抑菌圈直径值判定丁酸梭菌对大肠杆菌、金黄色葡萄球菌和沙门氏菌的抑制效果。抗氧化试验以1,1-二苯基-2-三硝基苯肼(DPPH)自由基、羟基自由基、超氧阴离子清除能力及还原力为依据。结果显示:上清液对3种指示菌的抑制作用最显著,均显著高于菌悬液、无细胞提取物和无菌蒸馏水(P<0.05);菌悬液和无细胞提取物对金黄色葡萄球菌、大肠杆菌和沙门氏菌的抑制作用次于上清液,显著高于无菌蒸馏水(P<0.05),且二者之间差异不显著(P>0.05)。丁酸梭菌上清液对DPPH自由基清除率显著高于菌悬液和无细胞提取物(P<0.05);上清液和菌悬液对羟基自由基清除率显著高于无细胞提取物(P<0.05);上清液对超氧阴离子自由基清除率显著高于菌悬液和无细胞提取物(P<0.05);还原力排序为上清液>菌悬液>无细胞提取物(P<0.05)。研究表明,丁酸梭菌对犊牛肠道致病菌具有一定的抑制作用,且具备抗氧化能力。The study investigates the antibacterial activity of Clostridium butyricum against intestinal pathogens(Staphylococcus aureus,Escherichia coli,Salmonella)in calves and its antioxidant properties.The inhibitory effect of Clostridiumbutyricumand its metabolites on Escherichiacoli,Staphylococcusaureusand Salmonellawas determined by the diameter of the inhibition zone using Oxford Cup method.The antioxidant test is based on the 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH)free radical,hydroxyl free radical,superoxide anion scavenging ability,and reducing power of Clostridiumbutyricum culture supernatant,bacterial suspension,and cell-free extract.The results showed that the Clostridium butyricum culture supernatant had the most significant inhibitory effect on Staphylococcus aureus,Escherichiacoli and Salmonella,which was significantly higher than those of bacterial suspension,cell-free extract and sterile distilled water(P<0.05).The inhibitory effect of bacterial suspension and cell-free extract on Staphylococcus aureus,Escherichiacoli and Salmonella was inferior to that of supernatant,which was significantly higher than that of sterile distilled water(P<0.05),and there was no significant difference between them(P>0.05).The DPPH free radical scavenging rate of Clostridiumbutyricumsupernatant was significantly higher than that of bacterial suspension and cell-free extract(P<0.05).The hydroxyl radical scavenging rate of supernatant and bacterial suspension was significantly higher than that of cell-free extract(P<0.05).The scavenging rate of supernatant was significantly higher than that of bacterial suspension and cell-free extract(P<0.05).The order of reducing power was supernatant>bacterial suspension>cell-free extract(P<0.05).The study indicates that Clostridium butyricum has a certain inhibitory effect on pathogenic bacteria in the intestinal tract of livestock and poultry,and has antioxidant capacity.
