摘 要:对比研究了3种悬钩子属植物(蔷薇梅、茅梅及覆盆子的脂溶性成分,并探究其体外药理活性。对3种植物的脂溶性进行提取,经气相色谱-质谱联用技术(GC-MS)分离和鉴定3种植物的脂溶性成分,用归一化法测定其相对百分含量,通过DPPH/ABTS^(+)清除自由基方法测定3种植物的抗氧化活性,采用Griess法检测细胞培养液中一氧化氮(NO)的含量,比较了3种植物的脂溶性成分的抗氧化活性和抗炎活性。共鉴定出104个化合物。蔷薇梅、茅梅和覆盆子中分别鉴定出54、51、51种化合物,分别占脂溶性成分总含量的68.33%、84.31%、80.27%;蔷薇梅、茅梅及覆盆子的清除DPPH自由基测定结果的IC50值分别6.67±2.14、0.07±0.02、3.78±1.04 mg/mL,清除ABTS+自由基测定结果的IC50值分别3.27±0.11、0.06±0.01、2.41±1.24 mg/mL,抗炎测定结果,与LPS模型组比较,蔷薇梅及覆盆子处理组的100、50、25μg/mL处理组细胞培养液中NO的含量均显著降低(P<0.01),茅梅处理组100、50μg/mL细胞培养液中NO的含量显著降低(P<0.01),且随着剂量的增加,NO含量有减少的趋势。三种悬钩子属植物中的成分主要是烷烃类、醇类、脂肪酸类成分,3种植物的成分及相对含量具有一定的差异性,显示出较好的抗氧化活性和抗炎活性。To comparatively study the liposoluble components of three Rubus species(Rubus rosifolius,Rubus parvifolius,and Rubus idaeus)and explore their pharmacological activities in vitro.The liposoluble components of the three plants were extracted and separated by gas chromatography-mass spectrometry(GC-MS).The relative percentage content of each component was determined by the normalization method.The antioxidant activity of the three plants was measured using DPPH/ABTS^(+)radical scavenging methods.Griess method was used to detect the content of nitric oxide(NO)in the cell culture medium to compare the antioxidant and anti-inflammatory activities of the liposoluble components of the three plants.A total of 104 compounds were identified.Specifically,54,51,and 51 compounds were identified in R.rosifolius,R.parvifolius,and R.idaeus,respectively,accounting for 68.33%,84.31%,and 80.27%of the total liposoluble content,respectively.The ICso values for DPPH radical scavenging were 6.67±2.14,0.07±0.02,and 3.78±1.04 mg/mL for R.rosifolius,R.parvifolius,and R.idaeus,respectively.The ICso values for ABTS radical scavenging were 3.27±0.11,0.06±0.01,and 2.41±1.24 mg/mL,respectively.For the anti-inflammatory activity,compared with the LPS model group,the NO content in the cell culture medium was significantly reduced in the 100,50,and 25μg/mL treatment groups of R.rosifolius and R.idaeus(P<0.01),and in the 100 and 50μg/mL treatment groups of R.parvifolius(P<0.01).Additionally,there was a tendency for the NO content to decrease with increasing dose.The main components of the three Rubus species are alkanes,alcohols,and fatty acids.The components and their relative contents vary among the three plants,but they all exhibit good antioxidant and anti-inflammatory activities.
