摘 要:目的运用高效液相色谱法结合响应面法优化续断中8种成分的提取工艺。方法利用超声提取,Box-Behnken Design设计,HPLC-UV对续断中三萜皂苷、环烯醚萜、挥发油这3类8种成分的含量进行测定。结果经单因素筛选,续断中8种活性成分的提取温度确定为30~40℃,超声频率确定为30~40k Hz,料液比确定为1∶15~1∶25(g∶m L),超声时长确定为30~35min。选取影响较大的因素超声起始温度、超声频率和料液比进行三因素三水平正交实验,综合5-羟甲基-2-呋喃甲醛、马钱苷酸、马钱子苷、当药苷、林生续断苷Ⅰ、续断苷B、续断苷A和川续断皂苷Ⅵ的提取率,确定优化的超声起始温度为34.84℃,超声频率为34.89k Hz,料液比为1∶22.04(g∶m L)。结论响应面法优化续断提取工艺结果较单因素筛选和未优化前的活性成分提取均有显著提高,表明建立的Box-Behnken Design模型稳定可靠,优化结果可以为续断药材和续断炮制品的合理开发及利用提供理论支持,同时可为其他中药材提取工艺的优化提供指导性意见。OBJECTIVE To optimize the extraction process of 8 components from Dipsaci Radix interrupts by high performance liquid chromatography(HPLC)combined with response surface method(RSM).METHODS Using ultrasonic extraction,Box-Behnken Design,HPLC-UV was used to determine the content of 8 components in 3 categories:iridoid,triterpene saponins and volatile oil.RESULTS After single factor screening,the extraction temperature of the 8 interrupted active ingredients was determined to be 30~40℃,the ultrasonic frequency was determined to be 30~40kHz,the solid-liquid ratio was determined to be 1∶15~1∶25,and the ultrasonic duration was determined to be 30~35 min.A three-factor,three-level orthogonal experiment was carried out by selecting the ratio of material to liquid with large influencing factors,initial ultrasonic temperature and ultrasonic frequency.After considering the extraction rate of 5-hydroxymethyl-2-furanoformaldehyde,loganic acid,loganin,sweroside,sylvestroside I,dipsanoside B,dipsanoside A and asperosaponinⅥcomprehensively,the optimized the ultrasonic starting temperature is 34.84℃,ultrasonic frequency is 34.89kHz and the solid-liquid ratio is 1∶22.04(g∶mL).CONCLUSION The results of response surface method optimized Dipsaci Radix extraction process were significantly higher than those of single factor screening and unoptimized extraction.The results show that the Box-Behnken Design model is stable and reliable,and the optimization results can provide theoretical support for the rational development and utilization of intermittent medicinal materials and intermittent products,and also provide guidance for the optimization of other Chinese medicinal materials extraction technology.
