三七总皂苷水解前后对血管新生及乳腺癌细胞的影响

摘  要:高温高压酸水解会对三七总皂苷(PNE)中人参皂苷类成分的结构产生影响,进一步探究该方法对PNE抗血管新生、细胞增殖和迁移的影响.采用物理和化学相结合的方法,即高温高压醋酸水解的方法,将PNE水解为人参次苷类成分;把制备的三七总皂苷水解产物(PNH)与PNE分别进行鸡胚绒毛尿囊膜(CAM)实验、细胞增殖实验和细胞划痕实验,分别考察其对血管新生和乳腺癌细胞MDA-MB-231的影响.实验结果表明,高温高压醋酸水解得到的PNH中含有人参皂苷Rk_(3)、20(S)-Rg_(3)、20(R)-Rg_(3)、Rk_(1)和Rg_(5),含量分别为4.70、9.22、5.29、23.52和6.68 mg/g;CAM实验中,PNE和PNH高剂量下对新生血管的抑制率分别达到26.23%和36.58%,都能抑制大血管中衍生的细支血管的生成;细胞增殖实验中,PNE和PNH都能够抑制乳腺癌细胞MDA-MB-231的增殖;细胞划痕实验中,PNE和PNH对乳腺癌细胞MDA-MB-231具有一定的细胞迁移抑制作用,PNH比PNE在相同浓度下显示出对细胞迁移的更大抑制作用.结果显示,PNH在相同浓度下表现出更强的血管新生抑制作用;PNE水解前后均能抑制乳腺癌细胞的增殖和迁移,而PNH活性更强;人参皂苷糖链经剪切变短后,生物活性和药理作用更为优异.The paper aims to investigate the effect of high-temperature and high-pressure acid hydrolysis on the structural components of ginsenosides in Panax notoginseng,and to further explore the effect of this method on angiogenesis,cell proliferation and migration.The combined physical and chemical methods,i.e.,high-temperature and high-pressure acetic acid hydrolysis,were used to hydrolyze Panax notoginseng saponins into ginseng secondary glycosides,and then the prepared Panax notoginseng saponins(PNH) and Panax notoginseng saponins(PNE) were subjected to chicken embryo chorioallantoic membrane(CAM) experiments,cell proliferation experiments and cell scratch experiments to investigate their effects on angiogenesis activity and MDA-MB-231 breast cancer cells,respectively.The experimental results showed that the PNH obtained by high temperature and high pressure acetic acid hydrolysis contained ginsenosides Rk_3,20(S)-Rg_3,20(R)-Rg_3,Rk_(1) and Rg_(5) with the contents of 4.70,9.22,5.29,23.52 and 6.68 mg/g,respectively,and the inhibition rates of PNE and PNH at high doses reached 26.23% and 36.58%,respectively in the CAM experiment.In the cell proliferation assay,PNE and PNH could inhibit the activity and proliferation of MDA-MB-231 breast cancer cells.In cell wound scratch assay,PNE and PNH inhibited cell migration of MDA-MB-231 breast cancer cells to some extent,and PNH showed greater inhibitory effect on cell migration than PNE at the same concentration.It can be concluded that these rare saponins showed stronger angiogenesis inhibition at the same concentration,the proliferation and migration of breast cancer cells were inhibited before and after the hydrolysis of Panax notoginseng saponins,and the PNH activity of the hydrolysate was stronger,and the biological activity and pharmacological effects of ginsenoside glycosides were better after shearing and shortening.
