摘 要:目的明确慢性间歇性低氧(chronic intermittent hypoxia,CIH)对睡眠觉醒过程的影响,进而探讨其在老年人睡眠障碍发生中的潜在作用。方法将8周龄雄性C57BL/6野生型小鼠随机分为CIH组和对照组,每组10只。CIH组进行持续4周的间歇低氧造模。造模结束后对小鼠进行在体多通道睡眠记录,观察睡眠觉醒量以及睡眠结构的改变。结果与对照组相比,持续4周的CIH造模可显著增加小鼠黑暗期非快速眼动(non-rapid eye movement sleep,NREM)睡眠总时长[(241.60±54.26)min vs(165.26±41.64)min,P<0.01]。睡眠结构分析显示,CIH小鼠在光照期睡眠过程中的觉醒片段数量明显增加[(294.10±55.00)vs(188.60±19.26),P<0.001],引起NREM睡眠片段平均时长明显缩短[(81.10±9.70)s vs(134.60±12.13)s,P<0.001],并且该片段化在黑暗期持续存在。结论CIH可导致睡眠觉醒过程紊乱,引起睡眠片段化改变并降低黑暗期的觉醒水平。Objective To investigate the effects of chronic intermittent hypoxia(CIH)on sleep-wake process and explore its potential role in sleep disturbances in the elderly.Methods The male C57BL/6 mice at the age of 8 weeks were randomly divided into CIH group and control group,with 10 ones in each group.The mice in the CIH group underwent CIH modeling in an intermittent hypoxic animal chamber for 4 weeks.After the modeling,the polysomnographic recordings were performed to observe the changes of total sleep-wake duration and sleep architecture.Results Compared with the control group,4-week CIH exposure significantly increased the total duration of non-rapid eye movement(NREM)sleep during the dark phase[(241.60±54.26)min vs(165.26±41.64)min,P<0.01].Sleep architecture analysis showed that the number of wake fragments in CIH mice increased significantly during the light phase[(294.10±55.00)vs(188.60±19.26),P<0.001].It resulted in a significantly shorter mean duration of NREM sleep fragments[(81.10±9.70)s vs(134.60±12.13)s,P<0.001],and these fragments persisted during the dark phase.Conclusion CIH can cause disturbance in the sleep wake process,which lead to sleep fragmentation and reduce the level of wakefulness during the dark phase.
