冠心舒通胶囊在脑损伤大鼠血浆、脑组织中的成分分析

摘  要:目的:定性分析冠心舒通胶囊在脑损伤大鼠血浆、脑组织中的移行成分。方法:采用线栓法复制大脑中动脉栓塞局灶性脑缺血模型(MCAO/R)。造模成功大鼠随机分为:模型对照组、冠心舒通胶囊4 g/kg组,另取正常大鼠设为假手术对照组、假手术+冠心舒通胶囊4 g/kg组。模型对照组、假手术对照组灌胃给予生理盐水,药物组给予等体积的冠心舒通药液,均为2次/d。连续灌胃5 d后采集全血并分离脑皮质,采用超高效液相色谱-电喷雾-四极杆-飞行时间-串联质谱技术(UPLC/Q-TOF MS)和气相色谱-质谱联用技术(GC-MS)法检测大鼠血浆、脑组织中的化学成分。结果:与假手术对照组相比,模型对照组神经功能评分、脑梗死体积及脑含水率均显著升高(P<0.01),提示造模成功;与模型对照组相比,冠心舒通4 g/kg组可明显降低MCAO/R大鼠神经功能缺损程度、脑梗死体积以及脑含水率(P<0.05或P<0.01)。两组冠心舒通治疗组中,大鼠血浆均可检测到冠心舒通10种主要的药效物质;其中,鞣花酸、丹酚酸B、丁香酚和龙脑还可在脑组织中被检测到;与假手术+冠心舒通4 g/kg组相比,冠心舒通4 g/kg组血浆成分色谱峰面积均降低,而脑组织成分色谱峰面积相当或轻度升高。结论:靶向入脑成分鞣花酸、丹酚酸B、丁香酚和龙脑可能是冠心舒通抗脑缺血损伤的关键药效物质。Objective:To qualitatively analyze the chemical components of Guanxinshutong(冠心舒通) Capsules(GXSTC) in the plasma and brain tissue of the rat model of cerebral ischemia-reperfusion injury. Methods:The rat model of MCAO/R was established by the suture method. After successful modeling, the rats were randomized into middle cerebral artery occlusion/reperfusion(MCAO/R),MCAO/R+GXSTC(4 g/kg) groups, normal rats were served as sham operation, sham operation+GXSTC(4 g/kg) groups. The MCAO/R and sham operation groups received saline, and the MCAO/R+GXSTC(4 g/kg) and sham operation+GXSTC(4 g/kg) groups were administrated with GXSTC twice a day. After 5 consecutive days of drug administration by gavage, blood and cerebral cortex samples were collected from each group, and then the chemical components in the plasma and brain tissue were detected by UPLC/Q-TOF MS and GC-MS,respectively. Results:Compared with the sham operation group, the MCAO/R group showed increased neurological deficit score, cerebral infarction volume, and water content rate in the brain(P<0.01),which suggested that the model was successfully established. Compared with those in the MCAO/R group, the neurological deficit score, cerebral infarction volume, and water content rate in the brain reduced in the MCAO/R+GXSTC group(P<0.05 or P<0.01). In the two groups with GXSTC,10 major components of GXSTC were detected in rat plasma. Among them, ellagic acid, salvianolic acid B,eugenol, and borneol were detected in the brain tissue. Compared with the sham operation+GXSTC group, the MCAO/R+GXSTC group showcased decreased peak areas of components in the plasma and similar or mildly increased content of components in the brain. Conclusions:Ellagic acid, salvianolic acid B,eugenol, and borneol may be the key pharmacodynamic components of GXSTC in treating cerebral ischemia-reperfusion injury.
