摘  要:目的通过一系列实验,探讨胰腺癌细胞源性外泌体(exo)对神经源细胞增殖及迁移功能的影响。方法通过聚合物沉淀法提取Panc02细胞的外泌体,并运用透射电镜、粒径分析及Western blot等方法对所提取的外泌体进行鉴定;将外泌体与大鼠雪旺细胞(RSC)和PC-12细胞共培养,实验分为4组,分别是磷酸缓冲盐溶液(PBS)组、50μg-exo组、100μg-exo组、200μg-exo组,通过CCK-8实验、平板克隆实验检测对细胞增殖能力的影响,通过Transwell实验、细胞划痕愈合实验检测对细胞迁移能力的影响。结果成功提取胰腺癌Panc02细胞的外泌体。CCK-8实验及平板克隆实验证明Panc02外泌体显著增强了神经源细胞的增殖能力。平板克隆实验中,加入外泌体后,RSC细胞的PBS组、50μg-exo组、100μg-exo组、200μg-exo组细胞增殖数分别为(24±6)、(39±5)、(68±4)和(56±7)个,差异有统计学意义(F=56.680,P<0.001);PC-12细胞的PBS组、50μg-exo组、100μg-exo组、200μg-exo组细胞增殖数分别为(52±6)、(122±4)、(138±3)和(101±5)个,差异有统计学意义(F=50.340,P<0.001)。Transwell实验结果显示,RSC细胞的PBS组、50μg-exo组、100μg-exo组、200μg-exo组细胞增殖数分别为(98±8)、(142±7)、(341±6)和(308±4)个,差异有统计学意义(F=79.940,P<0.001);PC-12细胞的PBS组、50μg-exo组、100μg-exo组、200μg-exo组细胞增殖数分别为(216±10)、(395±8)、(709±14)和(592±11)个,差异有统计学意义(F=95.280,P<0.001)。细胞划痕愈合实验结果显示,Panc02外泌体显著增强了神经源细胞的迁移能力(P<0.001)。结论胰腺癌细胞源性外泌体可以促进神经源细胞增殖和迁移能力。Objective A series of experiments were conducted to investigate the effect of pancreatic cancer cell-derived exosomes(exo)on the proliferation and migration of neurogenic cells.Methods The exosomes of Panc02 cells were extracted by polymer precipitation method,and identified by transmission electron microscopy,particle size analysis and Western blot.The exosome was co-cultured with rat Schwann cells(RSC)and PC-12 cells,and the experiment was divided into four groups,namely phosphate buffer saline(PBS)group,50μg-exo group,100μg-exo group,and 200μg-exo group.The effects of exosomes on cell proliferation were detected by CCK-8 assay and plate cloning assay.Transwell test and cell scratch healing test were used to detect the effect on cell migration ability.Results The exosomes of Panc02 cells of pancreatic cancer were extracted successfully.CCK-8 experiment and plate cloning experiment proved that Panc02 exosomes significantly enhanced the proliferation ability of neurogenic cells.In plate cloning experiment,after adding exosomes,the proliferation numbers of RSC cells in PBS,50μg-exo,100μg-exo and 200μg-exo groups were(24±6),(39±5),(68±4)and(56±7),respectively,and the differences were statistically significant(F=56.680,P<0.001);the proliferation numbers of PC-12 cells in PBS,50μg-exo,100μg-exo and 200μg-exo groups were(52±6),(122±4),(138±3)and(101±5),respectively,and the differences were statistically significant(F=50.340,P<0.001),the results of transwell experiment showed that there were(98±8),(142±7),(341±6)and(308±4)cells proliferating in the PBS,50μg-exo,100μg-exo and 200μg-exo groups of RSC cells,respectively,and the differences were statistically significant(F=79.940,P<0.001);there were(216±10),(395±8),(709±14)and(592±11)proliferation numbers of PC-12 cells in the PBS,50μg-exo,100μg-exo and 200μg-exo groups,respectively,and the differences were statistically significant(F=95.280,P<0.001).Cell scratch healing experiment also proved that Panc02 exosomes significantly enhanced the m
