摘  要:目的:研究白藜芦醇对阻塞性睡眠呼吸暂停低通气综合征(OSAHS)小鼠脾脏自噬水平的影响。方法:建立OSAHS C57BL/6小鼠动物模型,分为3组:常氧组、低氧组、白藜芦醇组(n=8)。Real-time PCR、Western blot和免疫组织化学染色(IHC)检测小鼠脾脏自噬相关蛋白微管相关蛋白1轻链3B(LC3B)、肌球蛋白样BCL2结合蛋白(Beclin1)和自噬相关基因5(ATG5)mRNA和蛋白的表达。结果:低氧组小鼠脾脏LC3B、Beclin1和ATG5 mRNA的表达较低于常氧组(P<0.01、P<0.01、P<0.001),白藜芦醇上调低氧组的LC3B和ATG5 mRNA表达水平(P<0.05、P<0.001),而不能明显上调Beclin1的表达(P>0.05)。低氧组小鼠脾脏LC3B、Beclin1、ATG5蛋白的表达表达较低于常氧组(P<0.05、P<0.05、P<0.01),白藜芦醇可上调低氧组的LC3B、ATG5表达水平(分别为P<0.05、P<0.01),而不能上调Beclin1的表达(P>0.05)。IHC染色显示低氧组小鼠脾脏LC3B、Beclin1、ATG5的表达低于常氧组(P<0.01、P<0.01、P<0.001),白藜芦醇可提高低氧组LC3B、ATG5的表达(P<0.05、P<0.01),不能上调Beclin1的表达(P>0.05)。结论:低氧抑制小鼠脾脏的自噬水平;白藜芦醇可上调低氧下小鼠的自噬水平,对Beclin1的作用不明显。Objective:To study the effects of hypoxia on the autophagy level in the spleen of mice with OSAHS.Methods:Mouse OSAHS hypoxia model was established in 24 C57BL/6 mice and the mice were divided into 3 groups:normoxic group,hypoxic group and resveratrol group(n=8).Real time PCR,Western blot and immunohistochemical staining(IHC)were used to detect the mRNA and protein expression of autophagy related proteins microtube associated protein 1 light chain 3 B(LC3B),myosin-like BCL2 inteacting protein(Beclin1)and autophagy related gene 5(ATG5)in mouse spleen respectively.Results:The mRNA expression of LC3B,Beclin1,and ATG5 in the hypoxic group was lower than that in the normoxic group(P<0.01,P<0.01,P<0.001),resveratrol upregulated the expression levels of LC3B and ATG5 mRNA in the hypoxic group(P<0.05,P<0.001),but did not upregulate the expression of Beclin1(P>0.05).The protein expression levels of LC3B,Beclin1 and ATG5 in the hypoxic group were lower than that in the normoxic group(P<0.05,P<0.05,P<0.01).Resveratrol upregulated the expression levels of LC3B and ATG5 in the hypoxic group(P<0.05,P<0.01),but did not upregulated the expression of Beclin1(P>0.05).IHC test showed that the expression level of LC3B,Beclin1 and ATG5 in the hypoxic group was lower than that in the normoxic group(P<0.01,P<0.01,P<0.001).Resveratrol increased the expression level of LC3B and ATG5 in the hypoxic group(P<0.05,P<0.01),but did not upregulate the expression of Beclin1(P>0.05).Conclusion:Hypoxia may inhibite the autophagy level of spleen in mice.Resvera-trol derivatives increases the autophagy level of mice under hypoxia condition,but has no significant effect on Beclin1 expression.
