摘 要:目的:探讨转录因子E2F1在头颈鳞状细胞癌(HNSC)组织中的表达、临床意义及预后相关性。方法:基于癌症基因组图谱(TCGA)数据库,分析E2F1在HNSC中的转录水平及其与HNSC患者生存率的关系和对HNSC的诊断价值。通过UALCAN数据库和GEPIA数据库分析E2F1在HNSC中表达水平及其与临床病理参数的相关性。通过CBioportal数据库分析E2F1的相关基因,进行KEGG通路富集分析及GO功能富集分析。纳入口腔鳞状细胞癌(OSCC)患者44例和正常口腔黏膜对照21例,免疫组织化学法(IHC)及RT-PCR法检测E2F1蛋白水平及mRNA水平的表达及其mRNA与临床病理的相关性。结果:TCGA数据显示,E2F1在癌组织中表达显著高于正常组织(P<0.01),与IHC及PCR检测结果一致(均P<0.01)。受试者工作特征曲线(ROC),显示曲线下面积(AUC)为0.966,在HNSC患者中E2F1的表达与预后呈正相关(P<0.05)。PCR结果显示,E2F1 mRNA在OSCC中呈高表达(P<0.01),不同分化程度(P<0.01),临床分期(P<0.05),有无淋巴结转移(P<0.05)具有统计学意义。与TCGA数据库中分析结果一致。不同年龄、性别组间差异无统计学意义(P>0.05),与TCGA的数据分析结果不一致。KEGG通路富集及GO功能分析结果显示,E2F1及其相关基因的生物过程主要涉及错配修复,细胞周期,DNA复制等过程。结论:E2F1的高表达可能与OSCC的发生、发展、转移密切相关。Objective:To investigate the expression,clinical significance and prognostic relevance of transcription factor E2F1 in head and neck squamous cell carcinoma(HNSC)tissues.Methods:Based on the cancer genome atlas(TCGA)database,the transcriptional level of E2F1 in HNSC,its relationship with diagnostic value and survival of HNSC patients were analyzed.Correlation of E2F1 expression levels in HNSC and its clinicopathological parameters were analyzed by UALCAN database and GEPIA database.Related genes of E2F1 were analyzed by CBioportal database,the coresponding results were studied by KEGG/GO enrichment analysis.The expression of E2F1 mRNA in 44 cases of oral squamous cell carcinoma(OSCC)and 21 controls of normal oral mucosa tissue was detected by immunohistochemistry and PCR.Results:TCGA data revealed that E2F1 expression in HNSC was significantly higher than in normal tissue(P<0.01),which was consistent with the results of IHC and PCR(both P<0.01).ROC showed that the AUC was 0.966,E2F1 expression was positively correlated with the prognosis of HNSC patients(P<0.05).PCR analysis showed that E2F1 mRNA level was high in OSCC and corelated with the differetation,clinical stage and limph metastasis(P<0.05),and not with the age and gender(P>0.05).Meanwhile,KEGG/GO enrichment analysis showed that biological processes of E2F1 and its related genes were mainly involved in mismatch repair,cell cycle,DNA replication and other processes.Conclusion:The high expression of E2F1 may be closely related to the occurrence,development and metastasis of HNSC.
