摘 要:目的应用非靶向代谢组学技术研究不同产地三叶青(TH)叶中的化学成分,筛选差异代谢物,为提高TH资源开发利用等提供理论依据。方法应用超高效液相色谱-串联质谱联用仪技术对闽选1号、临桂、天峨产地的TH叶进行非靶向代谢组学分析,采用主成分分析(PCA)、正交偏最小二乘判别分析(OPLS-DA)和聚类热图等方法,以变量投影重要度(VIP)值(VIP≥1)、显著性差异P值(以P<0.05为差异有统计学意义)和差异倍数(FC)值(FC≥2和FC<0.05)为指标,进行代谢物差异性分析。结果鉴定并筛选出3个产地TH叶中显著性差异代谢物36种,主要为黄酮及其苷类和酚酸类化合物,其中闽选1号、临桂和天峨样品分别有11、6、13个代谢物相对含量明显较高。结论基于超高效液相色谱-串联质谱联用仪的非靶向代谢组学技术鉴定了3个产地来源的TH叶代谢物,筛选出36种具有显著性差异的代谢物,为提高TH资源开发利用等提供理论依据。Objective To study the chemical composition of tetrastigma hemsleyanum(TH)leaves from different origins by non-targeted metabolomics technology,screen differential metabolites,and provide theoretical basis for improving the development and utilization of TH resources.Methods Utra-high performance liquid chromatography tandem mass spectrometry technique was used to analyze the non-targeted metabolomics of TH leaves from Minxuan number 1,Lingui and Tiane.principal component analysis(PCA),orthogonal partial least square-discriminant analysis(OPLS-DA)and cluster heat map were used.The variable importance in the projection(VIP)value(VIP≥1),the significant difference P value(P<0.05 was considered statistically significant)and the fold change(FC)value(FC≥2 and FC<0.05)were used as indicators to analyze the metabolite differences.Results A total of 36 significant differential metabolites were identified in the leaves of TH from the three habitats,mainly flavonoids,their glycosides and phenolic acids.Among them,11,6 and 13 metabolites were significantly higher in Minxuan number 1,Lingui and Tian'e samples,respectively.Conclusion Based on the non-targeted metabolomics technology of utra-high performance liquid chromatography tandem mass spectrometry,TH leaf metabolites from 3 origin were identified and 36 metabolites with significant differences were screened out,which provided theoretical basis for improving the development and utilization of TH resources.
