灰树花醇提物体外抗氧化及降血糖活性分析

摘  要:为研究灰树花醇提物的抗氧化及降血糖活性,以灰树花为原料,制备灰树花70%醇提物,并测定了醇提物中多酚、三萜、总糖和蛋白质的含量,通过DPPH自由基、ABTS自由基的清除能力和总还原能力测定灰树花醇提物的抗氧化能力,利用α-葡萄糖苷酶和α-淀粉酶的抑制作用评价灰树花醇提物的降血糖活性。此外,用高浓度胰岛素构建HepG2细胞胰岛素抵抗模型(IR-HepG2),进一步研究灰树花醇提物的抗氧化及降血糖活性。结果表明,灰树花70%醇提物中主要以蛋白质为主,其含量为504.14 mg·g^(-1);其对DPPH和ABTS自由基均有良好的清除作用,且IC_(50)分别为634.67μg·mL^(-1)和101.51μg·mL^(-1);灰树花醇提物还可以抑制α-葡萄糖苷酶和α-淀粉酶的活性,且IC_(50)分别为254.17μg·mL^(-1)和297.9μg·mL^(-1)。细胞试验结果表明,与模型组相比,灰树花醇提物能提高IR-HepG2细胞对葡萄糖的消耗,并促进细胞内糖原合成;经灰树花醇提物干预后,GSH-Px、CAT和SOD活力分别提高44.35%、138.73%、23.89%;MDA与ROS水平分别下降59.75%、25.06%。综上,灰树花醇提物具有良好的体外抗氧化及降血糖作用,能调节IR-HepG2细胞内抗氧化酶水平,改善氧化应激,从而改善HepG2细胞胰岛素抵抗。该研究结果可为灰树花在降血糖营养膳食方面的应用提供理论依据。In order to study the antioxidant and hypoglycemic activities of the alcohol extract of Grifolia grifolia,the 70%alcohol extract from G.grifolia was prepared as raw material,and the contents of polyphenols,triterpenoids,total sugars and proteins in the alcohol extract were determined.The antioxidant capacity of the alcohol extract was determined by the scavenging ability of DPPH and ABTS free radicals and the total reducing ability.The inhibitory effects ofα-glucosidase andα-amylase were used to evaluate the hypoglycemic activity of the alcohol extract from G.frondosa.In addition,insulin resistance model of HepG2 cells(IR-HepG2)was constructed with high insulin concentration to further study the antioxidant and hypoglycemic activities of the alcohol extract from G.frondosa.The results showed that protein was the main content of 70%alcohol extract of G.grifolia,and its content was 504.14 mg·g^(-1).It had good scavenging effect on DPPH and ABTS free radicals,and IC 50 was 634.67μg·mL^(-1) and 101.51μg·mL^(-1),respectively.The activity ofα-glucosidase andα-amylase was inhibited by the alcohol extract,and the IC 50 was 254.17μg·mL^(-1) and 297.9μg·mL^(-1),respectively.The cell experiments result showed that compared with the model group,the alcohol extract from G.frondosa could increase the glucose consumption of IR-HepG2 cells and promote the synthesis of glycogen.The activity of GSH-Px,CAT and SOD increased by 44.35%,138.73%and 23.89%,respectively.MDA and ROS levels decreased by 59.75%and 25.06%,respectively.In summary,the alcohol extract from G.frondosa has good antioxidant and hypoglycemic effects in vitro,can regulate the antioxidant oxidase level in IR-HepG2 cells,improve oxidative stress,and thus improve insulin resistance of HepG2 cells.The result in this study can provide theoretical basis for G.frondosa in hypoglycemic nutrition diet.
