摘  要:为探究植物激素对灵芝深层发酵的影响,研究了生长素对深层发酵灵芝菌丝生长和总三萜含量的影响。菌丝体总三萜含量在生长素浓度为10μmol·L^(-1)和50μmol·L^(-1)时显著高于对照组,而生长素浓度高于500μmol·L^(-1)时显著抑制菌丝体生长,当浓度为500μmol·L^(-1)时菌丝体絮状突起较长且粗,而当浓度为1 000μmol·L^(-1)时絮状突起较密且细。响应面分析结果显示当生长素浓度为19.5μmol·L^(-1),在发酵第5.6 d添加时,总三萜含量可达最大(1.322 g·100^(-1)·g^(-1))。经qRT-PCR检测得知,相比于对照组,在19.5μmol·L^(-1)生长素诱导下,灵芝菌丝体三萜合成途径中的3-羟基-3-甲基戊二酰辅酶A还原酶基因(HMGR)表达量显著降低,角鲨烯合成酶基因(SQS)表达量显著提高;而在50μmol·L^(-1)生长素诱导下菌丝体HMGR基因表达量显著降低。表明一定浓度的生长素可显著促进灵芝深层发酵中总三萜的生物合成,有助于灵芝三萜的进一步研究与应用。In order to investigate the effect of plant hormones on deep fermentation of Ganoderma lucidum,the effects of auxin on mycelial growth and total triterpene content of G.lucidum were investigated.The total triterpene content in mycelia of G.lucidum was significantly higher than that of the control group at auxin concentrations of 10μmol·L^(-1) and 50μmol·L^(-1),while the growth of mycelia was significantly inhibited when auxin concentration was higher than 500μmol·L^(-1).The morphology of floccus of mycelium was longer and thicker when auxin concentration was 500μmol·L^(-1),while it was dense and fine at 1000μmol·L^(-1).The response surface analysis showed that when the auxin concentration was 19.5μmol·L^(-1) and the addition time was 5.6 d,the total triterpene content reached its maximum(1.322 g·100-1·g^(-1)).According to qRT-PCR detection,compared to the control group,under the induction of auxin which concentration was 19.5μmol·L^(-1) the expression level of HMGR gene of G.lucidum mycelia in the triterpene synthetic pathway was significantly reduced,while the expression level of SQS gene was significantly increased.However,under the induction of auxin which concentration was 50μmol·L^(-1) only,the expression level of HMGR gene in mycelia was significantly reduced.The results indicates that a certain concentration of auxin can significantly promote the biosynthesis of total triterpene in G.lucidum deep fermentation,which is helpful for further research and application of triterpenes in G.lucidum.
