摘 要:目的:探讨不同浓度的鼓槌石斛乙醇提取物(DCLA)、鼓槌石斛水提物(DCLW)、流苏石斛乙醇提取物(DFHA)、流苏石斛水提物(DFHW)对秀丽隐杆线虫α-突触核蛋白聚集的抑制作用。方法:以秀丽隐杆线虫为模型生物,实验分为空白组、左旋多巴阳性药组和石斛提取物组,检测0.1、1、10、100 mg/mL的DCLA、DCLW、DFHA、DFHW的黄色荧光蛋白荧光强度,检测其抗α-突触核蛋白聚集作用。结果:与空白组相比,DCLA、DCLW、DFHA、DFHW均对α-突触核蛋白聚集显示出显著抑制作用,且在各实验组中,10 mg/mL均为最佳抑制浓度,其抑制活性由大到小为DCLA>DFHA>DCLW>DFHW。从不同溶剂提取方式来看,乙醇提取物比水提物显示出更明显的抑制活性,可能是生物碱类和黄酮类成分发挥的作用。结论:鼓槌石斛和流苏石斛水提物及醇提物均有一定的神经保护作用,10 mg/mL DCLA抗α-突触核蛋白聚集作用最强,石斛中的生物碱类和黄酮类可能是神经保护的活性成分。【Objective】To study the inhibitory effects of ethanol extract of Dendrobium chrysotoxum(DCLA),water extract of Dendrobium chrysotoxum(DCLW),ethanol extract of Dendrobium fimbriatum(DFHA),and water extract of Dendrobium fimbriatum(DFHW)on Caenorhabditis elegans’α-synapse aggregation in different concentrations.【Method】Using C.elegans as model organism,the experiment was divided into blank group,levodopa positive drug group and Dendrobium extract group.The fluorescence intensity of yellow fluorescent protein of DCLA,DCLW,DFHA and DFHW of 0.1 mg/mL,1 mg/mL,10 mg/mL and 100 mg/mL were detected.The anti-α-synaptonucleoprotein aggregation was detected.【Result】Compared with the blank group,DCLA,DCLW,DFHA,and DFHW showed significant inhibitory effects onα-synuclein aggregation,and in all experimental groups,10 mg/mL was the best inhibitory concentration,and their inhibitory activities were DCLA>DFHA>DCLW>DFHW in descending order.From the different solvent extracts,the ethanol extract showed more obvious inhibitory activity than water extract,which may be the role of alkaloids and flavonoids.【Conclusion】Both the aqueous and alcoholic extracts of Dendrobium drumstick and Dendrobium frutescens showed some neuroprotective effects,and 10 mg/mL DCLA had the strongest effect againstα-synuclein aggregation.The alkaloids and flavonoids in DCLA may be the active components of neuroprotective effects.
