摘  要:为明确苜蓿花叶病毒(Alfalfa mosaic virus,AMV)和白三叶草花叶病毒(White clover mosaic virus,WCMV)复合侵染黄花烟草后对绿原酸、总酚和类黄酮3种酚类物质的影响,设置单独接种AMV、WCMV以及复合接种AMV和WCMV(3∶1)侵染黄花烟草处理,以接种PBS缓冲液的黄花烟草为阴性对照(CK),观察接种后6、18、30 d时黄花烟草的发病症状并进行DAS-ELISA检测,同时在接种病毒后3、6、9、12、15、18、21、24、27、30 d时取黄花烟草叶片测定绿原酸、总酚和类黄酮含量(质量分数)。结果表明,黄花烟草中两种病毒复合侵染的症状表现重于单独侵染,表现为植株矮化,叶片皱缩、褪绿;在接种病毒后,AMV和WCMV两种病毒复合侵染的平均复制增殖率(30.68%和28.24%)低于单独接种后的平均复制增殖率(32.19%和30.27%);AMV和WCMV复合接种黄花烟草后,两种病毒复合侵染的黄花烟草叶片中绿原酸、总酚和类黄酮含量明显高于两种病毒单独侵染的处理。3种物质含量分别在接种病毒后30、27、30 d时达到最大值,分别为558.98、177.52和33.98μg·g^(-1)FW。两种病毒复合侵染的黄花烟草叶片中绿原酸、总酚和类黄酮含量达到最大值时与AMV、WCMV单独侵染和CK相比,分别提高98.28%、85.90%和107.76%,103.73%、42.78%和137.25%,39.03%、40.93%和63.84%,且差异均达到显著水平。表明两种病毒复合侵染后黄花烟草中绿原酸、总酚和类黄酮的产生与AMV和WCMV复合侵染胁迫存在相关性。To clarify the effect of co-infection with alfalfa mosaic virus(AMV)and white clover mosaic virus(WCMV)on three phenolic substances(chlorogenic acid,total phenols and flavonoids)in Nicotiana rustica,N.rustica inoculated with AMV and WCMV separately and combined in a ratio of 3∶1 was set as the treatment,while N.rustica inoculated with PBS buffer alone served as the negative control.Symptoms of N.rustica were observed and DAS-ELISA was performed 6,18 and 30 days after inoculation.At the same time,the leaves of N.rustica were collected 3,6,9,12,15,18,21,24,27 and 30 days after inoculation with viruses to determine the contents(mass fractions)of chlorogenic acid,total phenols and flavonoids.The results showed that the symptoms of N.rustica after co-infection with the two viruses were more severe than after single infection,showing plant dwarfism,leaf shrinkage and chlorosis.The average reproduction rate after co-inoculation with AMV and WCMV(30.68%and 28.24%)was lower than that after single inoculations(32.19%and 30.27%).After co-inoculation with AMV and WCMV,the contents of chlorogenic acid,total phenols and flavonoids in leaves of N.rustica infected were significantly higher than those inoculated with the single virus.The contents of the three substances reached maximum values of 558.98,177.52 and 33.98μg·g^(-1)FW at 30,27 and 30 d after inoculation,respectively.The maximum contents of chlorogenic acid,total phenolic and flavonoids in leaves of N.rustica after co-infection increased by 98.28%,85.90%and 107.76%,103.73%,42.78%and 137.25%,39.03%,40.93%and 63.84%compared to single infection with AMV,WCMV and CK and the differences were significant.The results showed that the production of chlorogenic acid,total phenols and flavonoids in N.rustica after co-infection with the two viruses was correlated with the co-infection stress of AMV and WCMV.
