基于网络药理学探讨中药三七治疗绒毛膜下血肿的作用机制

摘  要:目的基于网络药理学探讨中药三七治疗绒毛膜下血肿的作用机制。方法通过检索TCMSP、PubChem和SwissTargetPrediciton数据库得到三七的作用靶点;在DrugBank、GeneCards、OMIM和PharmGKB数据库检索绒毛膜下血肿(绒毛膜下出血)的疾病相关靶点;将筛选出的三七作用靶点与绒毛膜下血肿疾病相关靶点取交集,作为三七治疗绒毛膜下血肿的潜在作用靶点。通过STRING数据库构建潜在作用靶点的蛋白互作(PPI)网络,并筛选关键靶点;对潜在作用靶点进行基因本体论(GO)功能及京都基因与基因组百科全书(KEGG)通路富集分析,构建药物-化合物-疾病-靶点网络。结果获得三七治疗绒毛膜下血肿的潜在作用靶点26个;筛选得到雌激素受体1(ESR1)、信号转导与转录激活因子3(STAT3)、雄激素受体(AR)、孕激素受体(PGR)、成纤维细胞生长因子(FGF2)等关键靶点。潜在作用靶点GO功能富集分析得到82项生物途径(BP)条目、14项细胞组分(CC)条目、30项分子功能(MF)条目;KEGG通路富集分析共得到21条信号通路。结论三七可能通过ESR1、STAT3、AR、PGR、FGF2等关键靶点,作用于癌症通路、钙信号通路、神经活性配体-受体相互作用等主要信号通路,进而影响绒毛膜下血肿的核受体介导的类固醇激素信号通路、胞质钙离子浓度的正调节等生理过程,发挥治疗绒毛膜下血肿的作用。Objective To explore the mechanism of Panax Notoginseng in the treatment of subchorionic hematoma based on network pharmacology.Methods The action targets of Panax Notoginseng were obtained by searching the databases of TCMSP,PubChem and SwissTargetPrediction,and the disease-related targets of subchorionic hematoma(or subchorionic hemorrhage)were searched in the databases of DrugBank,GeneCards,OMIM and PharmGKB.The selected targets of Panax Notoginseng were intersected with the disease-related targets of subchorionic hematoma,which were used as potential targets for the treatment of subchorionic hematoma.The protein-protein interaction(PPI)network of potential targets was constructed by STRING database,and the key targets were screened;the potential targets were analyzed by GO function and KEGG pathway enrichment analysis,and the drug-compound-disease-target network was constructed.Results A total of 26 potential targets of Panax Notoginseng in the treatment of subchorionic hematoma were obtained,and key targets such as ESR1,STAT3,AR,PGR and FGF2 were screened.A total of 82 biological process(BP)items,14 molecular function(MF)items and 30 cellular component(CC)items were obtained by GO functional enrichment analysis of potential targets,and 21 signal pathways were obtained by KEGG pathway enrichment analysis.Conclusion Panax Notoginseng may act on the main signal pathways such as pathways in cancer,calcium signaling pathway,neuroactive ligand-receptor interaction through the key targets such as ESR1,STAT3,AR,PGR and FGF2,and then affect the physiological processes such as nuclear receptor-mediated steroid hormone signaling pathway,positive regulation of cytosolic calcium ion concentration and signal transduction in subchorionic hematoma,and play a role in the treatment of subchorionic hematoma.
