摘 要:目的 建立一套基于Image J的像素分析方法对薄层色谱法进行定量分析;借助人工智能自主开发软件简化Image J的像素分析流程;以茶叶中咖啡因的含量测定为模型,验证该方法的可行性。方法 先以图像处理软件Image J为工具建立像素分析方法,再通过ChatGPT,以人机交互命令的方式生成并优化新软件(TLC-Scan),实现Image J像素分析流程的自动化。最后以茶叶提取液为模型样品,验证这种基于TLC-Scan的薄层色谱定量方法的实用性。结果 ChatGPT生成的TLC-Scan软件可以快速直接给出目标斑点的积分密度作为定量参数,且灵敏度比Image J方法显著提高,该方法用于外标两点法测定茶叶提取液中咖啡因的含量时,表现出了较高的精密度、加样回收率和良好的重现性。结论 本文借助人工智能自主开发的软件及其相应的像素分析法可以快速获取薄层色谱像素中的积分密度信息,实现薄层色谱的定量分析,从而使薄层色谱定量分析摆脱对薄层扫描仪的依赖,尤其适合在设备条件不足情况下进行快速定量分析。OBJECTIVE To establish an image analysis method based on Image J for quantitative analysis of thin-layer chromatography(TLC);develop a software by artificial intelligence(AI)to practice the above method;to determine the caffeine content in tea using the method based on the software.METHODS First,the image processing software Image J was used as a tool to establish the image analysis method,and then with the help of ChatGPT,the new software(TLC-Scan)was generated and optimized by the way of human-computer interaction to achieve the automation of the image analysis process of Image J.Finally,the tea extract was used as a model sample to verify the practicality of this TLC-Scan-based quantitative method.RESULTS The TLC-Scan software generated by ChatGPT can quickly and directly give the integral density of the target spots as a quantitative parameter,and the sensitivity is significantly higher than that of the Image J method,which showed high precision,high spiked recoveries and good reproducibility in the determination of caffeine in tea extract by the external standard two-spot method.CONCLUSION The software developed by AI and the related pixel analysis method can quickly acquire the integral density information of TLC images to realize the quantitative analysis,which enables the quantitative analysis of TLC to get rid of the dependence on thin-layer scanners,and is especially suitable for the rapid quantitative analysis without sufficient equipment.
