摘 要:本试验旨在寻求分离纯化尖叶胡枝子总黄酮的最优大孔树脂,并优化其工艺参数,探究其体外抗氧化活性,以期提高黄酮产量。其中D-101型大孔树脂能较好地分离尖叶胡枝子总黄酮,且以总黄酮得量考察了上样流速、洗脱液(乙醇)浓度及流速对D-101树脂吸附性能的影响。结果表明:由正交试验确定最佳工艺参数为:洗脱液80%乙醇、进样速度1 mL/min、洗脱速度1.5 mL/min。经D-101大孔树脂精制后,尖叶胡枝子总黄酮纯度为52.21%,是原液(20.45%)纯度的2.55倍。尖叶胡枝子黄酮对羟自由基及超氧阴离子的清除率分别为98.5%和100%。本试验可为尖叶胡枝子作为植物源抗氧化剂应用于畜禽养殖生产实践提供参考依据。The purpose of this study was to find the optimal macroporous resin and its process parameters for the isolation and purification of total flavonoids from Lespedeza hedysaroides and to explore its antioxidant activity in vitro,in order to increase the flavonoid yield.Among them,D-101 macroporous resin was a resin material that could better separate the total flavonoids of Lespedeza hedysaroides.The effects of sample loading flow rate,eluent(ethanol)concentration and flow rate on the adsorption performance of D-101 resin were investigated by the total flavonoid yield,and the optimal process parameters were determined by orthogonal experiments:80%ethanol for eluent,1 mL/min injection rate and 1.5 mL/min for elution rate.After refining with D-101 macroporous resin,the purity of total flavonoids was 52.21%,which was 2.55 times that of the original solution(20.45%).The scavenging rates of hydroxyl radical and superoxide anion were 98.5%and 100%,respectively.This experiment could provide a reference for the application of Lespedeza hedysaroides as a plant-derived antioxidant in livestock and poultry breeding.
