摘 要:本试验旨在分离、纯化菊科药用植物款冬(Tussilago farfara L.)内生真菌,并研究其次级代谢产物的抗氧化活性。通过组织分离、平板划线等分离纯化方法,得到款冬根、茎、叶内生真菌32株,通过测定总还原力、DPPH自由基(DPPH·)清除率和羟自由基(·OH)清除率来探讨真菌发酵物的抗氧化活性。32株款冬内生真菌发酵产物中,菌株KD-18发酵产物乙酸乙酯萃取部位对DPPH·清除活性较强,当样品质量浓度为1mg/mL时,清除率为90.86%;KD-8发酵产物乙酸乙酯萃取部位对·OH清除活性较强,当样品质量浓度为1mg/mL时,清除率为89.32%;KD-8发酵产物乙酸乙酯萃取部位总还原力较强,在0~0.2mg/mL浓度范围内总还原力与对照品VC相当。结果显示,多数样品表现出一定抗氧化活性,其中KD-18乙酸乙酯萃取部位的发酵产物对DPPH·的清除率超过90%,表现出较强的抗氧化活性。药用植物款冬内生真菌和次级代谢产物的种类均较丰富,部分内生真菌代谢产物抗氧化活性较强,为开发利用药用植物款冬内生真菌资源提供了依据。The purpose of this experiment was to isolate and purify endophytic fungi from the medicinal plant Tussilago farfarfara L.in the Asteraceae family,and study the antioxidant activity of its secondary metabolites.32 strains of Tussilago farfara L.root,stem and leaf endophytic fungi were obtained by tissue separation,plate marking and other separation and purification methods,and the antioxidant activities of the fermentation products of the endophytic fungi of Tussilago farfara L.were preliminarily determined by using DPPH·radical clearance,hydroxyl radical clearance and total reducing force.Among the fermentation products of 32 strains of endophytic fungi from Tussilago farfara L.,DPPH·scavenging activity of the ethyl acetate extract of the fermentation product of strain KD-18 was better,and the DPPH·scavenging rate was 90.86%when the sample concentration was 1 mg/mL.KD-8 fermentation product ethyl acetate extraction site presented a higher·OH removal activity with a scavenging rate of 89.32%at a concentration of 1 mg/mL;total reducibility of the ethyl acetate extracted from the KD-8 fermentation product was strong,and within the concentration range of 0-0.2 mg/mL,the total reducibility was comparable to that of the VC control.DPPH·free radical content in the fermented product of KD-18 ethyl acetate extract exceeded 90%,demonstrating strong antioxidant activity.The endophytic fungi from Tussilago farfara L.and secondary metabolites are abundant,and some of the metabolites present good pharmacological activity,which can provide the basis for the comprehensive development and utilization of the endophytic fungi from Tussilago farfara L..
