灰树花多糖对胃溃疡大鼠胃肠菌群和氧化损伤的影响

摘  要:目的探讨灰树花多糖对胃溃疡大鼠胃肠菌群与氧化应激的影响。方法采用“乙酸灌胃法”制备50只胃溃疡模型大鼠,按随机数表法分为观察组与对照组各25只,观察组给予灰树花多糖提取物0.972 g/(kg·d)灌胃,对照组给予奥美拉唑1.04 mg/(kg·d)灌胃,干预前与干预8周后分别采用16S rRNA测序分析大鼠胃肠菌群,采集静脉血分析氧化应激指标与胃肠激素指标。结果干预后观察组胃肠菌群Ace指数为(373.63±59.82)、Chao指数为(1152.72±114.70)、Shannon指数为(7.53±0.45),均高于对照组的(315.72±63.64)、(1314.12±123.84)、(6.93±0.58),差异有统计学意义(P<0.05);干预后观察组克雷伯菌属、大肠杆菌-志贺氏菌属、阿克曼菌属丰度降低,乳杆菌属丰度升高,且变化幅度大于对照组,差异有统计学意义(P<0.05);观察组干预后超氧化物歧化酶水平为(46.17±2.14)mol/L、谷胱甘肽过氧化酶水平为(147.26±4.65)μmol/mL,均高于对照组的(43.85±2.25)mol/L、(133.69±5.27)μmol/mL,丙二醛水平为(8.55±0.67)mmol/L,低于对照组的(9.26±0.58)mmol/L,差异有统计学意义(P<0.05);观察组干预后胃泌素水平为(202.57±9.58)pg/mL、生长抑素水平为(236.97±8.59)pg/mL、胃动素水平为(223.35±20.74)pg/mL,均低于对照组的(223.73±5.83)pg/mL、(245.36±7.35)pg/mL、(239.64±21.47)pg/mL,差异有统计学意义(P<0.05)。结论灰树花多糖可有效调节胃溃疡大鼠胃肠菌群微生态平衡,抑制氧化应激,改善胃肠激素水平。Objective To investigate the effects of polysaccharides from Grifola frondosa on gastrointestinal flora and oxidative stress in rats with gastric ulcers.Methods 50 gastric ulcer model rats were prepared using the"acetic acid gavage method".They were randomly divided into an observation group and a control group,with 25 rats in each group.The observation group was given 0.972 g/(kg·d)of polysaccharide extract from Grifola frondosa by gavage,while the control group was given 1.04 mg/(kg·d)of omeprazole by gavage.Before and after 8 weeks of intervention,16S rRNA sequencing was used to analyze the gut microbiota of the rats,and venous blood was collected to analyze oxidative stress and gastrointestinal hormone indicators.Results After intervention,the Ace index,Chao index,and Shannon index of the gastrointestinal microbiota in the observation group were(373.63±59.82),(1152.72±114.70),and(7.53±0.45),all of which were higher than those in the control group(315.72±63.64,1314.12±123.84,6.93±0.58)(P<0.05);After intervention,the abundance of Klebsiella,Escherichia coli Shigella,and Akkermansia in the observation group decreased,while the abundance of Lactobacillus increased,and the magnitude of the change was greater than that in the control group(P<0.05);after intervention,the levels of superoxide dismutase and glutathione peroxidase in the observation group were(46.17±2.14)mol/L and(147.26±4.65)μmol/mL,respectively,which were higher than those in the control group(43.85±2.25)mol/L and(133.69±5.27)μmol/mL.The level of malondialdehyde was(8.55±0.67)mmol/L,which was lower than that in the control group(9.26±0.58)mmol/L(P<0.05);after intervention,the levels of gastrin,somatostatin,and motilin in the observation group were(202.57±9.58)pg/mL,(236.97±8.59)pg/mL,and(223.35±20.74)pg/mL,respectively,all lower than those in the control group(223.73±5.83)pg/mL,(245.36±7.35)pg/mL,and(239.64±21.47)pg/mL(P<0.05).Conclusion Polysaccharide from Grifola frondosa can effectively regulate the microbial balance of gastr
