摘 要:目的研究M2型巨噬细胞及其相关关键基因在食管鳞癌(esophageal squamous cell carcinoma,ESCC)中的浸润或表达水平,并评估其对ESCC患者预后的影响。方法从TCGA数据库获得ESCC基因表达矩阵,基于Rstudio利用免疫浸润CIBERSORT包评估ESCC组织中M2型巨噬细胞浸润水平,并通过Limma包进行差异分析鉴定出与M2型巨噬细胞相关的关键基因,最后采用Survival包分析评估M2型巨噬细胞及其相关关键基因对ESCC患者预后的影响。结果在ESCC组织中,22种免疫细胞的浸润水平存在差异,其中M2型巨噬细胞的浸润水平相对较高,并且与较差的生存率显著相关。FOLR2作为关键基因,在ESCC组织中高表达,并与病理分级、临床分期、TP53突变以及预后密切相关。单细胞数据分析提示FOLR2主要在巨噬细胞中表达。结论M2型巨噬细胞可能在ESCC进展中扮演着重要角色,FOLR2或可作为ESCC有效的生物标志物和潜在治疗靶点。Objective To investigate the infiltration or expression levels of M2 macrophages and their related key genes in esophageal squamous cell carcinoma(ESCC),and to assess their impact on the prognosis of ESCC patients.Methods The ESCC gene expression matrix was obtained from the TCGA database.Basingon Rstudio,the infiltration level of M2 macrophages in ESCC tissues was assessed with the immune infiltration CIBERSORT package,and the key genes related to M2 macrophages were identified through difference analysis with the Limma package.Finally,the influence of M2 macrophages infiltration level and related keys on the prognosis of ESCC patients was evaluated by survⅣal package.Results Studies have shown that there are differences in the infiltration levels of 22 types of immune cells in ESCC tissue,among which M2 macrophages have relatively higher infiltration levels and are significantly correlated with poorer survival rates.FOLR2,as a key gene,is highly expressed in ESCC tissues and is closely related to pathological grading,clinical staging,TP53 mutation and poor prognosis.Single-cell data analysis suggests that FOLR2 is mainly expressed in macrophages.Conclusion M2 macrophages play an important role in ESCC studies,and FOLR2 can be used as an effective biomarker and potential therapeutic target for ESCC.
