摘  要:目的通过生物信息学分析的方法,探究不同疗法对银屑病皮损基因表达的影响及相互作用。方法在GEO数据库中检索银屑病皮损及银屑病治疗相关基因数据,对数据集进行质量控制和预处理后筛选差异基因(DEGs)。通过GO和KEGG等工具对DEGs进行分析。结果治疗组共发现1036个DEGs,银屑病组共发现194个DEGs。功能分析表明,DEGs主要与角化过程、有丝分裂、趋化作用等过程有关;不同疗法在白细胞介素17(IL-17)通路、角化过程等方面有协同作用。结论从基因表达的层面,单克隆抗体尤其是以依那西普为代表的肿瘤坏死因子α(TNF-α)抗体以及IL-17抗体在银屑病治疗中表现出更高的适应性。Objective To investigate the effect and interactions of different therapies on gene expression in psoriasis based on the bioinformatics analysis. Methods GEO database was searched for the gene datasets of psoriasis skin lesion and psoriasis the-rapies,and quality control and pretreatment were performed for the datasets to obtain differentially expressed genes(DEGs).Gene ontology(GO)and Kyoto Encyclopedia of Genes and Genomes analyses were performed for the DEGs.Results There were 1036 DEGs in the therapy group and 194 DEGs in the psoriasis group.The GO functional enrichment analysis showed that the DEGs were mainly associated with keratinization,mitosis,and chemotaxis,and a synergistic effect was observed between different therapies in terms of the interleukin-17(IL-17)pathway and keratinization. Conclusion From the view of gene expression,mo-noclonal antibodies,especially the tumor necrosis factor-α(TNF-α)antibodies represented by etanercept and the IL-17 inhibitors,show a higher level of adaptability in the treatment of psoriasis.
