培元抗癌汤醇提物通过circ _SLIT3/NF-κB轴对HuH7人肝癌细胞侵袭、迁移及凋亡的影响

摘　　要：目的探讨培元抗癌汤醇提物对HuH7人肝癌细胞侵袭、迁移及凋亡的影响及其作用机制。方法实验设2组,细胞组的HuH7人肝癌细胞常规培养,药物组的HuH7人肝癌细胞加入培元抗癌汤醇提物200 mg/mL培养,Transwell实验检测2组HuH7人肝癌细胞侵袭及迁移情况,Western blot法检测2组细胞中B细胞淋巴瘤-2(Bcl-2)、Bcl-2相关X蛋白(Bax)、半胱氨酸-天冬氨酸蛋白酶-3(Caspase-3)、核因子-κB(NF-κB)蛋白表达情况,PCR法检测2组细胞中circ_SLIT3表达情况。结果药物组HuH7人肝癌细胞侵袭数目和迁移数目均明显少于细胞组(P均<0.05);药物组细胞中Bcl-2蛋白相对表达量和circ_SLIT3相对表达量均明显低于细胞组(P<0.05),Bax、Caspase-3、NF-κB蛋白相对表达量均明显高于细胞组(P均<0.05)。结论培元抗癌汤醇提物可能通过激活circ_SLIT3/NF-κB通路上调促凋亡蛋白表达,下调抗凋亡蛋白表达,从而抑制肝癌细胞的侵袭和迁移,促进细胞凋亡。Objective It is to investigate the effects and mechanisms of the alcoholic extract of Peiyuan Anti-cancer decoction on the invasion,migration and apoptosis of hepatocellular carcinoma cells HuH7.Methods Two groups were set up in the experiment,HuH7 human hepatocellular carcinoma cells in the cell group were cultured routinely,HuH7 human hepatocellular carcinoma cells in the medicated group were cultured with alcohol extract of Peiyuan Anti-cancer decoction 200 mg/mL.The cell invasion and migration of HuH7 human hepatocellular carcinoma cells of the two groups were detected by ranswell assay,the protein expressions of Bcl-2,Bax,Caspase-3 and nuclear factor-κB(NF-κB)in the cells of the two groups were detected by Western blot assay,and the expression of circ_SLIT3 in the cells of the two groups was detected by PCR.Results The numbers of invasion and migration of HuH7 cells in the medicated group were significantly lower than those in the cell group(all P<0.05);the relative expressions of Bcl-2 protein and circ_SLIT3 in the cells of the medicated group were significantly lower than those in the cell group(all P<0.05),and the relative expressions of Bax,Caspase-3,and NF-κB proteins were significantly higher than those in the cell group(all P<0.05).Conclusion The alcoholic extract of Peiyuan Anti-cancer decoction may significantly inhibit the invasion and migration of hepatocellular carcinoma cells and promote apoptosis via activating the circ_SLIT3/NF-κB pathway.
