摘 要:研究芍药汤(Shaoyao decoction,SYD)的抗氧化活性以及对H_(2)O_(2)诱导的人克隆结肠腺癌(Caco-2)细胞氧化损伤的保护作用。通过DPPH、ABTS、羟自由基清除试验以及FRAP法评价SYD提取物的体外抗氧化活力;筛选SYD对Caco-2细胞的无毒范围,并建立H_(2)O_(2)诱导的Caco-2细胞氧化损伤模型,检测SYD对氧化损伤细胞活力以及氧化应激指标的影响,评价SYD对Caco-2细胞氧化损伤的保护作用。结果显示,在一定范围内,SYD对DPPH、ABTS、羟自由基的清除能力以及对Fe^(3+)还原能力随浓度的升高而升高,EC_(50)分别为2.953μg/mL、1.492μg/mL和10.360μg/mL。SYD能显著提高H_(2)O_(2)诱导的氧化损伤Caco-2细胞活力,并显著抑制氧化损伤细胞NO的升高,增强SOD活力以及GSH的含量。结果表明SYD具有良好的抗氧化活性,能够有效缓解H_(2)O_(2)导致的Caco-2细胞氧化损伤。To study the antioxidant activity of Shaoyao decoction(SYD)and its protective effect against H_(2)O_(2)-induced oxidative damage in Caco-2 cells,the in vitro antioxidant activity of SYD extract was determined by DPPH,ABTS,hydroxyl radical scavenging assay and FRAP assay.The toxicity range of SYD on Caco-2 cells was screened,and a model of H_(2)O_(2)-induced oxidative damage in Caco-2 cells was established to detect the effects of SYD on the viability of oxidatively damaged cells as well as on oxidative stress indexes to evaluate the protective effect of SYD on oxidative damage in Caco-2 cells.The results showed that the scavenging ability of SYD for DPPH,ABTS,and hydroxyl radical,as well as its ability to reduce Fe^(3+),increased with the concentrations within a certain range,and the EC_(50)s were 2.953μg/mL,1.492μg/mL,and 10.360μg/mL,respectively.SYD significantly increased the cell viability of the oxidatively damaged Caco-2 cells induced by H_(2)O_(2),and significantly inhibited the elevation of NO,enhanced the SOD activity and GSH content in oxidatively damaged Caco-2 cells.The results indicated that SYD has good antioxidant activity and can effectively alleviate the oxidative damage of Caco-2 cells caused by H_(2)O_(2).
