摘 要:为探究姜黄(CurcumalongaL.)基因组密码子的偏好性使用模式,本研究通过CodonW1.4.4和Python程序对NCBI中下载的姜黄基因组68447条编码序列进行分析,利用中性绘图、ENC-plot、PR2-plot进行姜黄密码子偏好性分析,并对姜黄及其他6种生物密码子的偏好性进行比较。结果显示,姜黄基因组密码子不同位置上的GC含量表现为GC_(1)(53.77%)>GC_(3)(53.76%)>GC_(2)(42.87%),其中,RSCU>1的33个密码子以C或G结尾的有17个,表明姜黄基因组密码子末位碱基较偏好使用C/G;自然选择和突变压力是影响姜黄基因组密码子偏好性的主要因素。不同物种密码子偏好性比较发现姜黄密码子偏好性与洋葱差异最小。该研究结果不仅为姜黄基因组学的研究提供理论基础,也为姜黄的异源基因表达及应用提供指导。In order to explore the preference usage pattern of codons in Curcuma longa L.genome.The complete turmeric genome sequence was downloaded from NCBI and its structure was analyzed.CodonW1.4.4 and Python programs were used to analyze the 68447 coding se-quences of ginger genome,and important parameters such as GC_(1),GC_(2),GC_(3),RSCU,and ENc were obtained.Neutral mapping,ENC-plot,PR2-plot,and optimal codon analysis were performed.The results showed that the GC content at different positions of the turmeric genome codons was GC_(1)(53.77%)>GC_(3)(53.76%)>GC_(2)(42.87%).Among them,17 of the 33 codons with RSCU>1 ended in C or G,indicating that the base at the end of the turmeric genome codon preferred to use C/G;the codon usage bias of ginger genome is mainly affected by natural selection and mutation pressure factors.Through comparison,it was found that the codon preference of turmeric was different from that of Saccharomyces cerevisiae and other species,among which the difference with onion was the smallest.The results not only provide a theoretical basis for the study of turmeric genomics,but also provide theoretical guidance for the heterologous gene expression of turmeric.
