摘 要:为了解犬细小病毒临床分离株VP2基因的遗传进化规律。本研究采集1例疑患细小病毒病的犬粪便样品,通过提取样品中的病毒基因组,采用PCR扩增及测序,将序列与不同种的细小病毒VP2基因序列进行核苷酸和氨基酸比对,并进行遗传进化分析。结果显示,该粪便样品CPV/LS-01中扩增出的VP2核苷酸序列为CPV-2c型,与参考毒株MF467242(CPV-2c)的核苷酸同源性最高为99.9%,与M38245(CPV-2)同源性较低(98.8%),与其它参考毒株的同源性为99.0%~99.7%。对VP2蛋白中氨基酸变异情况分析显示,CPV/LS-01样品中的VP2蛋白的I447M位氨基酸位点出现了CPV-2c型细小病毒的独特突变。本研究中所鉴定的犬腹泻粪便中的细小病毒属于CPV-2c型,与目前河南分离毒株(ON322797)VP2中具有相同的突变位点(A5G和I447M),该研究为犬细小病毒流行趋势的研究提供了一定的理论依据。To understand the genetic evolution of VP2 gene of canine parvovirus clinical isolates.The faces of 1 dog with canine parvovirus disease was collected.Then extracted the viral DNA,amplified VP2 gene by PCR and sequenced;then gave the homology,genetic evolution and amino acid variation analysis with the VP2 gene sequences of isolated strains and those of other strains belong to the same genus.The results showed that the genetype of strain CPV/LS-01 is CPV-2c,which has the highest nucleotide homology with MF467242(CPV-2c)was 99.9%and the lowest nucleotide homology with M38245(CPV-2)was 98.8%,and homology with other reference strains were between 99.0%and 99.7%.The amino acid variation analysis of VP2 protein showed that the mutation at site I447 of VP2 protein in CPV/LS-01 was different from other CPV-2c strains.The CPV identified in this study belongs to CPV-2c type and has the same locus mutation(A5G and I447M)in VP2 of the current strain isolated from Henan Province(ON322797),which provides a theoretical basis for the study of the epi-demic trend of canine parvovirus.
