双硫死亡相关基因作为常见恶性肿瘤预后指标及治疗靶点的研究进展

摘　　要：人体细胞通过自我毁灭,以维持生物稳态和保护身体免受致病物质侵害,这一过程可归为代谢性细胞死亡(regulated cell death,RCD)。最近的研究发现了一种全新的代谢相关RCD形式,名为双硫死亡。双硫死亡是细胞内二硫化物积累增强了还原型烟酰胺腺嘌呤二核苷酸磷酸(nicotinamide adenine dinucleotide phosphate,NADPH)的还原能力,以抵消双硫死亡和维持细胞存活,与现有的其他形式的细胞死亡不同。在此之后,越来越多的研究人员试图确定双硫死亡与肿瘤之间的关系,为癌症治疗开拓新的可能性。因此,本文系统地阐述了双硫死亡相关分子表达和细胞代谢过程以及双硫死亡相关的肿瘤信号通路,重点梳理发现过程中的表达及机理,概述双硫死亡与癌症之间的关系,同时,汇总双硫死亡相关基因可能作为常见恶性肿瘤中的生物标志物及治疗靶点,为临床运用双流死亡相关基因参数影响恶性肿瘤诊疗提供认知性参考。The process of self-destruction of human cells to maintain biological homeostasis and protect the body from pathogenic substances is called regulated cell death(RCD).Recent studies have found a new form of metabolically related RCD,called disulfide death.Disulfide death is the accumulation of intracellular disulfide that enhances the reduction ability of nicotinamide adenine dinucleotide phosphate(NADPH)to counteract disulfide death and maintain cell survival,unlike other forms of cell death that exist today.Since then,more and more researchers have tried to determine the relationship between disulfide death and tumors,opening up new possibilities for cancer treatment.Therefore,this paper systematically expounds the expression of disulfide death-related molecules,cell metabolic processes and disulfide death-related tumor signaling pathways.It combs the expression and mechanism in the key discovery process,summarizes the relationship between disulfide death and cancer.Meanwhile,it also summarizes that disulfide death-related genes may be used as biomarkers and therapeutic targets in common malignant tumors,and provides a cognitive reference for clinical application of disulfide death-related gene parameters affecting the diagnosis and treatment of malignant tumors.
