壳聚糖/三七抗菌止血海绵的制备及性能研究

摘　　要：壳聚糖具有抗菌止血、无毒促凝血等特点,被广泛应用于止血海绵的制备。本文以壳聚糖和三七为基本原料,通过冷冻干燥法制备壳聚糖/三七复合海绵,使海绵兼具壳聚糖和三七的抗菌止血性能。通过红外光谱、扫描电子显微镜、抑菌实验、溶血凝血和体外细胞实验等测试分析技术和方法,对海绵的理化性能和生物相容性进行检测。结果表明:相较于壳聚糖海绵,壳聚糖/三七复合海绵具有更大的孔隙且孔洞分布均匀,对金黄色葡萄球菌和大肠杆菌具有显著的抑菌效果,对红细胞具有较好的粘附作用,具有更好的止血效果,同时体外细胞培养实验证明海绵没有明显的细胞毒性。综上所述,壳聚糖/三七抗菌止血海绵具有良好的抗菌止血性能和生物相容性,是一种优良的止血材料。Chitosan is widely used in the preparation of hemostatic sponge with its antibacterial,hemostatic,non-toxic and coagulant promoting features.In this paper,chitosan/notoginseng composite sponge was prepared by freeze-drying method with chitosan and notoginseng as the basic raw materials,so that the sponge had the antibacterial and hemostatic properties of both chitosan and notoginseng.The physicochemical properties and biocompatibility of sponges were analyzed and evaluated using infrared spectroscopy(FT-IR),scanning electron microscopy(SEM),antibacterial tests,hemolysis and coagulation assays and vitro cell experiment.The results indicated that compared to chitosan sponges alone,the chitosan/PNS sponges exhibited bigger and uniformly distributed pores,demonstrating significant antibacterial effects against Staphylococcus aureus and Escherichia coli.They also showed improved adhesion to red blood cells and superior hemostatic performance.Meanwhile,the vitro cell experiment proved that the sponges have no obvious cytotoxicity.In summary,chitosan/notoginseng antibacterial hemostatic sponge is an excellent hemostatic material for it has good antibacterial,hemostatic performance and biocompatibility.
