摘  要:研究甘木通叶片总黄酮(Total flavonoids in Clematis filamentosa Dunn leaves,CLTF)提取工艺优化及其功能活性.采用亚硝酸钠-硝酸铝-氢氧化钠法测定CLTF含量;采用微波辅助乙醇提取法,通过单因素和响应面试验,优化CLTF的提取工艺.结果表明,CLTF的最佳提取工艺为:微波时间61 s、乙醇浓度61%、液料比22∶1(mL·g^(-1))和微波功率300 W;CLTF的最佳提取率为(8.363±0.135)mg·g^(-1);CLTF清除DPPH自由基、羟基自由基和ABTS阳离子自由基的IC50值分别为0.040 5、0.032 6和0.041 6 mg·mL^(-1);CLTF抑制α-葡萄糖苷酶活性的IC50为0.002 1 mg·mL^(-1).综上可知,CLTF具有良好的抗氧化和降血糖活性,为甘木通叶片在食品、药品和保健品等方面的开发利用提供参考.The extraction process optimization and the functional activity evaluate of total flavonoids from Clematis filamentosa Dunn leaves(CLTF)were studied.The CLTF contents were quantified using the sodium nitrite-aluminum nitrate-sodium hydroxide colorimetric method.Microwave-assisted ethanol extraction was employed for the process parameters optimizing,through single-factor experiments and response surface methodology.The results showed that the optimal extraction conditions were determined as follows:microwave time of 61 s,ethanol concentration of 61%(v/v),liquid-to-material ratio of 22∶1(mL·g^(-1)),and microwave power of 300 W,yielding a maximum CLTF extraction rate of(8.363±0.135)mg·g^(-1).The antioxidant activities were evaluated through radical scavenging assays,with IC50 values of 0.0405,0.0326,and 0.0416 mg·mL^(-1) for DPPH,hydroxyl,and ABTS radicals,respectively.Furthermore,CLTF demonstratedα-glucosidase inhibitory activity with an IC50 value of 0.0021 mg·mL^(-1).These findings suggest that CLTF possesses antioxidant and hypoglycemic properties,highlighting its potential applications in foods,pharmaceuticals,and nutraceuticals.
