摘 要:为研究基于荧光标记技术使用荧光素显现潜血手印的可行性,探究其显现的最佳条件,选择两种常用荧光素(FITC荧光素和DCFH-DA荧光素)作为本次实验的主要研究对象,基于荧光标记技术显现潜血手印的原理,设计了使用荧光素显现非渗透性客体上潜血手印的方法及步骤。在此基础上,利用PBS缓冲液改变荧光素溶液pH环境,利用丙酮分别对两种荧光素溶液进行5倍、10倍、100倍的稀释,以探究手印的最佳显现条件。结果显示:利用FITC荧光素及DCFH-DA荧光素显现非渗透性客体上潜血手印的效果均较好。其中,FITC荧光素显现的最佳条件为:显现时间为10 min,pH环境值为9.35,浓度为原浓度;DCFH-DA荧光素显现的最佳条件为:显现时间为20 min,pH环境值为13.01,浓度为原浓度。因此,基于荧光标记技术使用荧光素显现非渗透性客体上的潜血手印是可行的,但还需对配制好的荧光素溶液的保存时间以及这种显现技术对血手印中DNA的破坏情况等进一步探究,综合分析其实际应用价值。In order to study the feasibility of using fl uorescein to develop latent blood fi ngerprints based on fl uorescent labeling technology and explore the optimal conditions for its development,we selected two cselected used fl uoresceins(FITC fl uorescein and DCFH-DA fl uorescein)as the main research objects of this experiment,to design the method and procedure of developing latent blood fi ngerprints on non-porous surfaces with fl uorescein,based on the principle of fl uorescent labeling technology.On this basis,PBS buffer was used to change the pH environment of fl uorescein solution,and acetone was used to dilute the two fl uorescein solutions by 5 times,10 times and 100 times,respectively,in order to explore the best developing conditions of latent blood fingerprints.The results showed that both FITC fluorescein and DCFH-DA fluorescein were effective in developing latent blood fi ngerprints on non-porous surfaces.The best conditions for FITC fl uorescein to develop latent blood fi ngerprints on non-porous surfaces are:the developing time being 10 minutes,the pH environment value being 9.35,and the concentration being the original concentration;The best conditions for DCFH-DA fl uorescein to develop latent blood fi ngerprints on non-porous surfaces are:the developing time being 20 minutes,a pH environment value being 13.01,and the concentration being the original concentration.Therefore,it is feasible for the fl uorescence labeling technology to develop latent blood fi ngerprints on non-porous surface,but it is still necessary to further explore the preservation time of prepared fl uorescein solution and the damage of DNA detection in blood fi ngerprints by this technology,and comprehensively analyze its practical application value.
