摘  要:目的观察左路升降散对慢性不可预见性轻度应激抑郁症模型大鼠行为和线粒体能量代谢的影响,明确左路升降散的抗抑郁作用并探究其作用机制。方法根据60只适应性饲养1周后雄性Wistar大鼠的行为学及体重情况,将其随机分为空白组、模型组、氟西汀组和左路升降散低、中、高剂量组,每组10只。除空白组外,其余各组均采用孤养和慢性不可预见性轻度应激法建立抑郁症模型,且在每天刺激后模型组灌胃蒸馏水,氟西汀组灌胃3.33 mg/kg的盐酸氟西汀,左路升降散高、中、低剂量组分别灌胃8 g/kg、6 g/kg、4 g/kg的左路升降散,持续35 d。每周称大鼠体重,实验结束进行行为学检测(旷场实验记录5 min内的运动距离、静止时间,新环境进食抑制实验记录进食潜伏期),HE染色观察大鼠海马组织神经元形态,酶联免疫吸附实验(ELISA)检测海马组织中ATP酶、琥珀酸脱氢酶(SDH)、细胞色素C氧化酶(COX)活性。结果实验第7,14,21,28,35天时,模型组大鼠体重均明显低于同期空白组(P均<0.05);实验第14,21,28,35天时,左路升降散低、中、高剂量组和氟西汀组大鼠体重均明显高于同期模型组(P均<0.05)。与空白组比较,模型组大鼠5 min内的运动距离明显缩短(P<0.05),静止时间、进食潜伏期均明显延长(P均<0.05);海马神经元排列疏松紊乱,部分神经元核固缩,神经纤维缠绕成结;海马组织中ATP酶、SDH、COX活力均明显降低(P均<0.05)。与模型组比较,左路升降散低、中、高剂量组和氟西汀组大鼠5 min内的运动距离均明显增加(P均<0.05),静止时间、进食潜伏期均明显缩短(P均<0.05);神经元损伤程度均减轻;海马组织中ATP酶、SDH、COX活力均明显升高(P均<0.05)。其中左路升降散高剂量组和氟西汀组神经元损伤减轻更明显,其余指标各药物组间比较差异均无统计学意义(P均>0.05)。结论左路升降散可明显改善抑郁症大鼠体重下降�Objective It is to observe the observe the effects of Zuolu Shengjiang decoction on the behavior and mitochondrial energy metabolism of model rats with depression induced by chronic unpredictable mild stimulation,to clarify the antidepressant effect of this medicne and to explore its mechanism of action.Methods Sixty male Wistar rats were randomly divided into blank group,model group,fluoxetine group and low-,medium-and high-dose groups of Zuolu Shengjiang decoction after 1 week of acclimating feeding according to their behavior and body weight,with 10 rats in each group.The rats of all groups except for the blank group were stimulated by solitary rearing and chronic unprdictable mild stress to establish models of depression.After each day’s stimulation,the model group was gavaged with distilled water,the fluoxetine group was gavaged with 3.33 mg/kg of fluoxetine hydrochloride,and the high-,medium-and low-dose groups of Zuolu Shengjiang decoction were gavaged with 8 g/kg,6 g/kg,and 4 g/kg of Zuolu Shengjiang decoction,respectively,all the groups were continuously treated for 35 days.The rats were weighed weekly,and at the end of the experiment,their behaviors including movement distance and resting time within 5 min in the open field test and feeding latency in the new environment feeding inhibition test were recorded,the morphology of neurons in hippocampal tissues of rats was observed by HE staining,and the activities of ATPase,succinate dehydrogenase(SDH),and cytochrome C oxidase(COX)in the hippocampal tissues were detected by enzyme-linked immunosorbent assay(ELISA).Results On the 7th,14th,21st,28th and 35th days of the experiment,the body weights of rats in the model group were significantly lower than those of rats in the blank group during the same period(all P<0.05);on the 14th,21st,28th and 35th days of the experiment,the body weights of rats in the low-,medium-and high-dose groups of Zuolu Shengjiang decoction and fluoxetine group were significantly higher than those of rats in the model group during
