摘 要:目的探讨唾液腺腺样囊性癌中VEGF(vascular endothelial growth factor,VEGF)表达。利用注射用低分子肝素钙溶液培养腺样囊性癌细胞SACC-83,观察其对VEGF和细胞增殖、侵袭和迁移的影响。方法免疫组织化学染色观察唾液腺及唾液腺腺样囊性癌中VEGF表达。采用10%胎牛血清-RPMI1640完全培养液将低分子量肝素注射液按照0 IU/mL、25 IU/mL、50 IU/mL、100 IU/mL、200 IU/mL梯度稀释,培养唾液腺腺样囊性癌SACC-83细胞。MTT增殖实验检测不同浓度低分子肝素对SACC-83细胞的作用,并将低分子肝素的半抑制浓度作为实验组(half maximal inhibitory concentration,IC50),SACC-83细胞作为对照组。Transwell侵袭实验和划痕实验,观察低分子肝素对SACC-83细胞侵袭和迁移的影响。Real-time PCR、Western blot检测两组细胞中VEGF表达。ELISA检测两组细胞培养液中VEGF含量。结果免疫组织化学染色显示正常唾液腺组织中VEGF呈低表达,唾液腺腺样囊性癌组织中VEGF呈高表达。MTT结果显示,SACC-83细胞培养72h,不同浓度低分子肝素对SACC-83均产生抑制作用,半抑制浓度IC50为49.34 IU/mL。Transwell侵袭和划痕实验显示,低分子肝素可明显抑制SACC-83细胞的侵袭和迁移。与对照组相比,低分子肝素组VEGF基因、蛋白表达明显降低,P<0.01。ELISA检测显示低分子肝素组细胞培养液中的VEGF表达降低,与对照组差异显著,P<0.01。结论低分子量肝素可能通过降低VEGF表达抑制SACC-83细胞的增殖、侵袭和迁移。Objective To investigate the expression of vascular endothelial growth factor(VEGF)in salivary gland adenoid cystic carcinoma and to observe the effects of low molecular weight heparin calcium injection on VEGF expression in the adenoid cystic carcinoma cell line SACC-83,as well as on cell proliferation,invasion,and migration.Methods The expression of VEGF in salivary gland and adenoid cystic carcinoma were assessed through immunohistochemical staining.The low molecular weight heparin for injection was diluted with 10%fetal bovine serum-RPMI1640 to a concentration of 0 IU/mL,25 IU/mL,50 IU/mL,100 IU/mL and 200 IU/mL.Subsequently,SACC-83 cells were cultured with different concentrations of low molecular weight heparin.The effect of low molecular weight heparin on SACC-83 cells was assessed by MTT proliferation assay.The half maximal inhibitory concentration(IC50)of low molecular weight heparin was determined.The experimental group consisted of SACC-83 cells treated with IC50 of low molecular weight heparin,while the control group comprised untreated SACC-83cells.The effects of low molecular weight heparin on invasion and migration of SACC-83 cells were assessed by transwell assay and scratch assay.The expression of the VEGF gene and protein was evaluated by Real-time PCR and Western blot.VEGF levels in the cell culture medium of both groups were measured via ELISA.Results Immunohistochemical staining revealed a decreased expression of VEGF in normal salivary gland tissues,whereas an increased expression of VEGF was observed in salivary gland adenoid cystic carcinoma.MTT results demonstrated that different concentrations of low molecular weight heparin effectively suppressed the growth of SACC-83 cells for a duration of 72 hours.The IC50 value of low molecular weight heparin was determined to be49.34 IU/mL.The invasion and migration of SACC-83 cells were inhibited by low molecular weight heparin,as demonstrated by transwell assay and scratch assay.Furthermore,a significant decrease in the expression of VEGF gene an
